过敏性腹泻小鼠大肠黏膜中T细胞亚群的表达与Th1/Th2型细胞因子的关系

摘　　要：目的 检测小鼠过敏性腹泻模型大肠黏膜中T细胞亚群的表达和血清中Th1型细胞因子（IFN-γ）、Th2型细胞因子（IL-4）和OVA-IgE水平,探讨腹泻小鼠大肠黏膜中T细胞亚群的表达特点与Th1/Th2型细胞因子的关系.方法 雌性BALB/c小鼠20只随机分成两组,即对照组和实验组.采用酶联免疫吸附法（ELISA）检测血清中IFN-γ,IL-4及OVA-IgE的水平,取大肠组织HE染色观察大肠组织病理改变,免疫组织化学染色观察T细胞亚群的表达情况.结果 与对照组相比,实验组小鼠非特异性炎症反应明显,上皮不规则排列,腺区存在大量白细胞（WBC）浸润,固有层可见大量嗜酸性粒细胞（EOS）浸润.CD3,CD4,CD8免疫组织化学染色显示,与对照组相比,肠黏膜固有层可见大量CD3＋,CD4＋,CD8＋细胞.实验组IL-4,OVA-IgE分泌水平明显升高,IFN-γ分泌水平和IFN-γ/IL-4比值显著降低（P〈0.01）.嗜酸粒细胞百分比与CD4＋/CD8＋比值呈显著正相关（P〈0.05）.结论 过敏性腹泻小鼠大肠黏膜局部CD4＋T细胞亚群明显增加,CD4/CD8细胞比例失衡,并与Th1/Th2型细胞因子向Th2型漂移及EOS浸润程度相关.Objective To establish a mouse model of allergic diarrhea and the proximal colon mucosa T cell subsets expression and the levels of Thl type cytokine （ IFN-γ）, Th2 type cytokine （ IL-4 ） and OVA-IgE in sera would be detected, then the relationship of T cell subsets and Th1/Th2 cytokines in allergic diarrhea would been analyzed. Methods Twenty female BALB/c mice were randomly divided into two groups ： control group and experimental group. The levels of IFN-γ, IL-4 and OVA-IgE in sera were detected by ELISA. Proximal colon was stained by HE and the pathological changes of colons in both groups were assessed. The expression of T cell subsets in colons of both groups were examined by immunohistochemistry. Results HE staining of proximal colon slices observed by light microscope showed that in experimental group of the colon mucosa epithelial obvious nonspecific inflammation reaction could be seen,the epithelial disorganized, the accumulation of WBC cells in the lamina propria of colon were significantly increased in experimental group than that of control group, and eosinophils infiltration were more evident in experimental group. Immunohistochemistry staining of colon sections showed that T cell subsets staining of experimental group were sinificantly positiver than that of control group. IL-4 and OVA-IgE levels in the sera of mice in experimental group were increased significantly than that in control group（ P 〈 0.01 ）. However,the IFN-γ and IFN-γ/IL-4 levels in the sera of mice in experimental group were decreased significantly than that in control group（P 〈 0.01）. The percentage of eosinophils was significantly positively related to CD4 ＋/CD8＋ ratio. Conclusion The expression of CD4 is predominate and CD4 ＋/CD8 ＋ ratio is imbanlance. CD4 cells may aggravate Th2-biased inflammation by inducing Thl/Th2 imbalance and are related to eosinophils infiltration.
