摘  要:目的研究洋葱水提取物和乙醇提取物对结肠癌细胞株凋亡及细胞周期的影响。方法采用水提取和乙醇提取洋葱后,用紫外可见分光光度仪检测提取物中黄酮类化合物含量。体外培养结肠癌细胞株(LoVo cells、SW480 cells、HT-29 cells、HCT-8 cells),采用含黄酮类化合物浓度40 mg/L和60 mg/L的提取物对其进行干预,流式细胞仪检测洋葱水提取物和乙醇提取物对结肠癌细胞株凋亡影响,检测含黄酮类化合物浓度为40 mg/L的水提取物和乙醇提取物对结肠癌细胞周期的影响。结果洋葱水提取物和乙醇提取物对结肠癌细胞株的凋亡具有剂量依赖性,其中水提取物引起SW480、HCT-8细胞株凋亡率较乙醇提取物高。而乙醇提取物引起HT-29细胞株凋亡率较水提取物高。洋葱提取物能将结肠癌细胞株的细胞周期阻滞在G1期,阻碍其向S期转换,以抑制细胞增殖。结论洋葱水提取物和乙醇提取物能够诱导结肠癌细胞株的凋亡,并能够对结肠癌细胞周期产生影响,其具体机制需进一步探讨。ObjectiveTo study the influence of water extract and ethanol extract apoptosis and cycle of colorectal caneer cells. MethodsAllium cepa was extracted by water and ethanol,the flavonoid content in extracts was detected by UV.Colorectal cancer cells(Lovo cells、SW480 cells、HT-29 cells、HCT-8 cells) were cultured in vitro,the apoptosis of cultured cells in different concentration of flavonoids(40、60mg/L)were detected by flow cytometry,the influence of cell cycle was tested with concentration of flavonoids 40mg/L. ResultsBoth the water extract and ethanol extract(detected by flow cytometry) could enhance the colorectal cancer cells' apoptosis rate and showed a dose dependence,the apoptosis rate induced by water extract was higher than ethanol extract in SW480 and HCT-8 cells,but the ethanol extract caused a higher apoptosis rate than water extract in HT-29 cells.Allium cepa extracts could block the cell cycle at G1 phase,hindered cells conversion to S phase then inhibited cell proliferation. ConclusionAllium cepa extracts can induce the apoptosis rate of colorectal cancer cells and has a notable influence on cell cycle.The mechanism need to be further studied.
