摘 要:目的观察并比较延胡索及其有效成分左旋延胡索乙素(l-tetrahydropalmatine,l-THP)对大鼠吗啡条件性位置偏爱(conditioned place preference,CPP)效应消退时间的影响。方法剂量递增法建立大鼠吗啡CPP模型,雄性SD大鼠颈背部注射吗啡和生理盐水训练10 d,在12 d确认CPP模型建立成功后,连续21 d给予延胡索2,1,0.5 g.kg-1水提液(分别含l-THP0.153,0.077,0.038 mg),以及l-THP 3.76,1.88,0.94 mg.kg-1灌胃治疗,观察各组大鼠吗啡CPP效应的消退时间。结果与生理盐水对照组比较,吗啡模型组大鼠在伴药箱停留的时间明显延长(P<0.05);治疗后,延胡索2,1g.kg-1水提液和l-THP 3.76,1.88 mg.kg-1组的CPP效应在用药6 d消退,而模型组、延胡索0.5 g.kg-1和l-THP0.94 mg.kg-1组在用药第2天消退。结论延胡索和l-THP均能加速大鼠吗啡CPP效应的消退,但含一倍量l-THP单体的延胡索中药相当于单独应用约24倍l-THP单体的效果。ObjectiveTo observe and compare effects on conditioned place preference(CPP) extinction in morphine-induced rats between Corylalis Yanhusuo and L-telra-hyoropalmatine. MethodsThe model of CPP was established by escalating doses of morphine in rats.Male SD rats were hypodermically injected with morphine and saline for 10 days.Different doses(2,1 and 0.5 g·kg-1) of Corydalis yanhusuo(pure l-THP:0.153,0.077 and 0.038mg·kg-1) were applied to treat the rats for 21 days when the model of CPP was validated on the 12 th day.Another group was treated with l-THP(3.76,1.88 and 0.94 mg·kg-1).The extinction time of each group was recorded. ResultsCompared with NS group,the prolonged detention time of model group was notably observed in drug-paired compartment(P0.05).Moreover,the reduction effects on CPP were observed in the groups of water extract(2,1 g·kg-1) and l-THP(3.76,1.88 mg·kg-1) subjected to medicine for 6 days(P0.05).However,the similar effects were found among the model group,Corydalis yanhusuo group(0.5 g·kg-1) and l-THP(0.94 mg·kg-1) group after 21-day treatments(P0.05). ConclusionCorydalis yanhusuo and l-THP can substantially accelerate the reduction effects in rats suffered from morphine.However,Corydalis yanhusuo is 24 times higher in efficacy than that of l-THP.
