荞麦黄酮复方制剂对2型糖尿病大鼠心血管病变的保护作用

摘　　要：目的探讨荞麦黄酮复方制剂对糖尿病心血管病变的保护作用。方法将SD大鼠70只通过高脂高热量饮食加小剂量链脲佐菌素腹腔注射建立大鼠2型糖尿病模型。观察空腹血糖(FBG)、放射免疫分析法测定血浆内皮素-1(ET-1)、胸主动脉细胞间黏附分子-1(ICAM-1)及胸主动脉血管细胞间黏附分子-1(VCAM-1)的含量,硝酸还原酶法测定血清一氧化氮(NO)含量,ELISA法检测血清血管内皮细胞生长因子(VEGF)含量。取胸主动环观察不同累积浓度的乙酰胆碱(Ach)对去甲肾上腺素(NE)引起的血管收缩的抑制率。结果 TFBCP能剂量依赖性抑制升高的FBG、血浆ET-1和血清VEGF;升高血清NO水平。模型组血管环对NE的收缩反应均明显强于正常对照组和H-TFBCP组。在相同Ach浓度下,模型组胸主动脉环对Ach的舒张作用明显弱于正常组及各TFBCP组。各TFBCP组能明显抑制糖尿病大鼠胸主动脉ICAM-1及VCAM-1表达。结论荞麦黄酮复方制剂能够保护糖尿病大鼠内皮依赖的血管舒张功能,具有抗动脉硬化作用。ObjectiveTo study the protective effect of the flavonoid of Buckwheat Compound Preparation（TFBCP） on cardiovascular complications in type 2 diabetes rats and its mechanism. MethodsThe SD rats were given high-fat diet,then were stimulated by the injection of small dosage of streptozocin to build diabetic model.The content of fasting blood glucose（FBG） was detected.Radioimmunoassay technique was used to measure ICAM-1 and VCAM-1 contents in thoracic aorta.The content of nitric oxide（NO） and vascular endothelial growth factor（VEGF） in serum were determined by nitrate reductase assay and ELISA assay,respectively.Thoracic aortic rings were used to detect the vascular activity. ResultsTFBCP reduced FBG,ET-1 and VEGF contents in a dose-dependent manner,improved the NO content.The contraction response induced by NE in model group was stronger than control group and H-TFBCP group.Under the same concentration,the relaxation of thoracic aorta in model group was weaker than control group and TFBCP groups.The expression of ICAM-1 and VCAM-1 could be obviously inhibited in TFBCP group compared with model group. ConclusionTFBCP has some protective effects on the endothelium of vessel in diabetes rats,and plays the role of anti-atherosclerosis.
