德国牧羊犬成纤维细胞培养及其生长特性研究

摘　　要：试验旨在探讨犬耳部和腹部皮肤成纤维细胞体外培养和纯化方法,为下一步犬体细胞核移植提供基础。结果表明,在组织块培养后的4d,耳组织周围开始有细胞出现,8～10d呈优势生长,2周后可长满细胞瓶。腹部皮肤组织则要6～7d方可见有细胞沿组织块周围爬出,10d左右可以在组织块周围形成成片细胞,20d左右可以长满整个细胞瓶。腹部皮肤成纤维细胞建系初期生长速度低于耳成纤维细胞,但传代至第4代时,两种细胞的生长曲线基本相同。For further studies in canine nuclear transfer,the culture and purification methods of canine ear and abdominal skin fibroblast cell were studied.The results showed the cells could be seen around ear tissue after 4 d culture and quick growth were seen about 8 to 10 d,after 2 weeks the bottle could be covered with cells.In abdominal skin group,cells could be seen after about 6 to 7 d and lots of cells around tissue appeared after about 10 d.The bottom of cell bottle would fully cover with cells after 20 d in abdominal skin group.The growth speed of abdominal skin fibroblast cells were lower than ear fibroblast cells in early stages of the cell lines establishment,but when cells grew to 4 passages,both the two cell lines had the same growth curve.
