不同治法方药对阿霉素肾病大鼠疗效比较以及与IL-10的相关性研究

摘　　要：目的:比较6种不同治法方药对肾病综合征大鼠的疗效,初步探讨其肾脏保护作用的可能机制,确立对肾病综合征大鼠的最佳治疗方药。方法:用阿霉素分次尾静脉注射法复制肾病综合征大鼠模型,实验动物随机分为8组,正常对照组、模型对照组、益气补肾组、收敛固涩组、清热利湿组、活血化瘀组、疏风通络组、综合治疗组。药物治疗组按6g生药材/200g体重给大鼠灌胃4周。测定尿蛋白、血清白蛋白、总蛋白、大鼠白细胞介素10的含量。结果:治疗4周后,益气补肾组的24h尿蛋白含量与模型组相比,显著性降低(P<0.05);血清中白蛋白含量显著性升高(P<0.05)。益气补肾组、疏风通络组、综合治疗组的IL-10含量与模型组相比,都显著性降低(P<0.05,P<0.01)。结论:在诸多治法方药中对肾病大鼠蛋白尿的疗效比较依次为益气补肾组>综合治疗组>活血化瘀组>疏风通络组>收敛固涩组,提示慢性肾小球疾病的治疗应重用益气补肾中药,益气补肾中药能够降低血清IL-10的水平,而使体液免疫的高反应状态得到调节。Objective：Compare the efficiency of six different prescriptions for nephrotic syndrome in rats,to investigate the possible mechanism of their renal protective function.And establish the best therapeutic prescription on rats model with adriamycin induced nephropathy.Methods：The rat model of nephropathy was established by injection of adriamycin through their tail vein.Experimental animals were randomly divided into 8 groups,normal group,control group,Yiqi Bushen group,Shoulian Guse group,Qingre Lishi group,Huoxue Huayu group,Shufeng Tongluo group and combined treatment group.Drug treatment group rats were administered for 4 weeks by 6g/200g raw drug and body weight proportion.Then the urinary protein,serum albumin,total protein and interleukin-10 content were detected.Results：Compared with the model group after treatment of 4 weeks,24h urine protein content in Yiqi Busheng group was decreased significantly（P〈0.05）;serum albumin content increased significantly（P〈0.05）.Compared with the model group,IL-10 content in Yiqi Bushen group,Shoulian Guse group and combined treatment group were decreased significantly（P〈0.05,P〈0.01）.Conclusion：In comparison with various therapeutics on nephropathy rat proteinuria efficacy,Yiqi Bushen was the best,followed by combined treatment group.The third was Huoxue Huayu group,the fourth Shufeng Tongluo group and the last Shoulian Guse group.It is suggested that chronic glomerular disease should be treated for reuse of Yiqi Bushen formula because it can reduce the levels of serum IL-10,and regulate the intensive humoral immune response.
