五派杨树多色荧光原位杂交分析(英文)

摘　　要：杨树染色体由一对较大的和17对较小的形态近似的染色体组成,联合利用18S-5.8S-25S rDNA,5SrDNA an拟南芥型端粒序列三种荧光探针分析大叶杨,胡杨,毛白杨二倍体,毛白杨三倍体,武黑一号,箭杆杨和小青杨七种五派杨树的分子核型.依据端粒信号测量染色体并根据不同杨树派别间rDNA定位染色体的不同区分杨树形态近似的17对染色体,并按降序排列各杨树的分子核型.分析表明,五派杨树中除胡杨外均由两个18S-5.8S-25SrDNA杂交位点,且位点位于较大的染色体上,而5S rDNA位点位于较小的染色体上,同时观察到七种杨树中均存在随体并与18S-5.8S-25S rDNA共定位于染色体端部.多色荧光原位杂交技术的利用为杨树的核型分析提供了更多的信息,有利于从分子水平区分杨树形态近似的染色体.The chromosomes（2n=2x=38） of Populus are composed of one pairs of large and 17 pairs small similar chromosomes at the morphological level.Three probes（18S-5.8S-25S rDNA,5S rDNA and Arabidposis-type telomere repeats） were located on the chromosomes of the seven Populus species which belong to the five Populus sections（Leuce,Aigeiros,Tacamahaca,Leucoides,Turanga）,by using the multicolor fluorescence in situ hybridization（M-FISH）.The rDNA locus and measuring data according to the chromosomes end displayed by the telomere hybridization fluorescence were applied to discriminate of the similar chromosomes and karyotype in decreasing order.The two 18S-5.8S-25S rDNA sits were observed in the Populus species except of P.euphratica where only one was identified.The major 18S-28S rDNA site was on the former chromosome.One 5S rDNA site was characterized in all seven.The satellites were observed in the seven Populus species and colocated with the 18S-28S rDNA sites.The multicolor FISH will be useful for identifying the chromosomes and karyotyping,and it can provide more information about difference among the species at the cytogenentic level.
