摘 要:目的:探讨蛹虫草乙酸乙酯提取物(GSCME)对高糖诱导人脐静脉内皮细胞(HUVECs)的衰老和细胞内活性氧的影响。方法:体外培养HUVECs,分3组:对照组(D-葡萄糖5.5mmol/L)、高糖组(D-葡萄糖40mmol/L)、GSCME组(GSCME 50μg/ml+D-葡萄糖40mmol/L),检测各组细胞增殖情况,SA-β-gal阳性衰老细胞,周期分布和细胞内活性氧(ROS)含量。结果:与对照组相比,高糖干预24、48、72h后细胞增殖活性显著下降[OD:48h:(0.599±0.062)比(0.375±0.013)],P均<0.01;高糖孵育48h后SA-β-gal阳性细胞率[(50.70±1.49)%比(89.63±0.69)%]、G0/G1期细胞百分率[(49.15±2.56)%比(76.59±3.58)%]、细胞内活性氧(ROS)水平[(1.03±0.15)比(3.60±0.36)]均明显增加,P均<0.01。与高糖组相比,GSCME孵育48h、72hHUVECs增殖率显著上升[48h:(0.375±0.013)比(0.424±0.023)],P均<0.05,孵育48h显著降低SA-β-gal阳性细胞率[(89.63±0.69)%比(65.81±1.50)%]、G0/G1期细胞百分率[(76.59±3.58)%比(52.70±5.64)%]及细胞内ROS水平[(3.60±0.36)比(1.67±0.21)],P<0.05~0.01。结论:蛹虫草提取物可延缓高糖诱导的人脐静脉内皮细胞衰老进程,降低细胞内活性氧从而减轻细胞氧化损伤可能是其作用的机制之一。Objective: To study influence of ethyl acetate extract of Cordyceps militaris(GSCME) on senescence of human umbilical vein endothelial cells(HUVECs) and reactive oxygen species(ROS) in cells induced by high glucose.Methods:HUVECs were cultured in vitro and divided into control group(cultured with D-glucose 5.5 mmol/L),high glucose group(cultured with D-glucose 40 mmol/L) and GSCME group(cultured with GSCME 50 μg/ml+D-glucose 40 mmol/L).HUVECs proliferation,SA-β-gal positive senescence cell,distribution of cell cycles and concentrations of ROS in cells were measured in each group.Results:Compared with control group,after incubated with high glucose for 24,48 and 72h,proliferation activity of HUVECs significantly decreased,P0.05 all];after incubated with high glucose for 48h,there were significant increase in SA-β-gal positive cell rate [(50.70±1.49)% vs.(89.63±0.69)%],percentage of G0/G1 period cells [(49.15±2.56)% vs.(76.59±3.58)%]and level of ROS [(1.03±0.15) vs.(3.60±0.36)],P0.01 all.Compared with high glucose group,HUVECs proliferation rate significantly increased after incubated with GSCME for 48h and 72h [48h:(0.375±0.013) vs.(0.424±0.023),]P0.05 both;after incubation with CME for 48h,there were significant decrease in SA-β-gal positive cell rate [(89.63±0.69)% vs.(65.81±1.50)%],percentage of G0/G1 period cells [(76.59±3.58)% vs.(52.70±5.64)%]and ROS level in cells [(3.60±0.36) vs.(1.67±0.21) ]in GSCME group,P0.05~0.01.Conclusion: GSCME can delay senescence progression of HUVECs induced by high glucose.One of its possible mechanisms may be ROS level in cell decrease,so it decreases oxidative damage of cells.
