摘 要:目的:研究莽草酸(SA)对血小板聚集性的影响及其机制,并研究其对凝血时间的影响。方法:采用比浊法测定血小板聚集功能,放射免疫法测定血小板TXB2 和血浆6ketoPGF1α水平,玻管法测定凝血时间。结果:SA 体外呈浓度依赖性抑制ADP、胶原诱导的兔血小板聚集,IC50 分别为9-25 ,3-56 m mol·L-1 ;在体内(ip) 抑制脑血栓模型鼠由ADP诱导的血小板聚集;SA促进大鼠血浆6ketoPGF1α的生成并延长小鼠凝血时间。结论:SA 对血小板聚集及凝血系统有显著的抑制作用。AIM: To study the inhibitory effects of shikimic acid (SA) on platelet aggregation and its mechanism, and the effect on coagulation time. METHODS: Methods of turbidimetry and radioimmunoassay were used to measure platelet aggregation and the levels of thromboxane B 2(TXB 2) and 6 keto PGF 1α . RESULTS: SA inhibited rabbit platelet aggregation induced by ADP and collagen in vitro with IC 50 of 9 25 and 3 56 mmol·L -1 , respectively. SA 25 and 50 mg·kg -1 ip for 3 d inhibited platelet aggregation induced by ADP in rats subjected to middle cerebral artery thrombosis(MCAT). SA was shown to increase rat plasma 6 keto PGF 1α and no significant effect on TXB 2 generation in PRP induced by ADP. SA 25, 50 and 100 mg·kg -1 im or iv elevated the blood coagulation time of mice. CONCLUSION: SA inhibited platelet aggregation and blood coagulation.
