摘  要:目的:研究冷冻解冻对大肠埃希菌超微结构的损伤和修复。方法:将正常的和经冷冻解冻损伤和修复后的大肠埃希菌作负染色和超薄切片,在电子显微镜下观察其超微结构的变化。结果:(1)冷冻解冻破坏大肠埃希菌的超微结构,细胞壁、细胞膜损伤,胞内有空泡形成;(2)修复后的大肠埃希菌,其超微结构基本恢复正常;(3)冰晶的形成、胞质浓缩产生的盐害是冷冻损伤的主要机理。结论:冷冻解冻可破坏大肠埃希菌的超微结构。Objective:To study the influence of freeze and thaw on damage and recovery of the ultra structure of E.coli cells.Methods:The changes of the ultra structure were observed under the electron-microscope by scanning the minus dyed section and ultra thin section of normal,freeze-thaw and recovered E.coli.Results:1.After freeze and thaw,the ultra structure of E.coli were changed with the cell wall and membrane damaged.The vacuole was formed in the cytoplasm;2.After cultivation,the ultra structure of E.coli was almost recovered.3.The ice-crystal and salt-harm formed by the concentration of the cytoplasm were the mechanisms of the freeze-injury.Conclusion:Freeze and thaw can damage the ultra structure of E.coli cells.
