温石棉的细胞转化效应及干预效应的研究

摘　　要：目的　探讨几种化合物对温石棉致人胚肺 (HEL)细胞转化效应的影响。方法　用柠檬酸铝、混合稀土或亚硒酸钠溶液浸泡温石棉 1h后 ,再将其与HEL细胞共同孵育 ,观察HEL细胞的转化灶数目。结果　温石棉可致HEL细胞发生形态转化 ,0 62 5、1 2 5、2 5、5 0 μg ml石棉所致细胞转化灶数分别为 ( 0 60± 0 5 5 ) 皿、( 1 2 0± 1 3 1) 皿、( 2 60± 1 14 ) 皿、( 3 60± 1 14 ) 皿 ,呈明显的剂量反应关系 ,其中 2 5、5 0 μg ml石棉组与对照组相比差异有显著性 ,表明温石棉可致HEL细胞发生形态转化。与未处理温石棉组相比 ,经 3种化合物预处理的温石棉所致HEL细胞转化灶数目均明显减少 ,其中混合稀土处理组的抑制率为 5 0 % ,差异有显著性。结论　用上述 3种化合物预处理温石棉 ,有可能减轻温石棉对人类的致癌危害性。Objective Toexploretheeffectsofseveralkindsofchemicalsonchrysotile inducedcell transformationinhumanembryolung (HEL)cells .Methods Afterbeingsoakedinaluminumcitrate ,mixedrare earthorsodiumselenitesolutionsforonehour,chrysotilewasincubatedwithHELcells.Then ,thenumberof focusofcelltransformationwasobserved .Results ChrysotilecouldcausemorphologicaltransformationofHEL cellsinadose dependentpattern ,with ( 0 60± 0 5 5 ) ,( 1 2 0± 1 3 1) ,( 2 60± 1 14 )and ( 3 60± 1 14 )foci ofHELcelltransformationperplateaftertreatmentwith 0 62 5 ,1 2 5 ,2 5and 5 0 μg mlofchrysotile, respectively,andtherewassignificantdifferencebetweenthegroupstreatedwith 2 5and 5 0 μg mlofchrysotile andcontrolgroup .ItindicatedthatchrysotilecouldcausemorphologicaltransformationofHELcells.Numberof focusofHELcelltransformationinducedbychrysotilethatwaspretreatedwiththeabove mentionedthreekindsof chemicalsreducedsignificantly ,ascomparedwiththatinthegroupwithoutpretreatmentofit.Itsinhibitoryrate was5 0 %inthegrouptreatedwithmixedrareearth ,withsignificantdifference .Conclusion Pretreatmentfor chrysotilewiththesethreekindsofchemicalscouldreduceitscarcinogenicity .
