摘  要:目的:研究聚甲基丙烯酸甲酯(Polymethyl methacrylate,PMMA)骨水泥微粒对MC3T3-E1成骨细胞株(osteoblast,OB)增殖、分化和功能的影响,以初步探究人工关节无菌性松动的发生机制。方法:采用MTT法检测细胞增殖的变化,对硝基苯磷酸法和考马斯亮兰法作碱性磷酸酶(ALP)比活性测定。结果:PMMA微粒在各浓度(2.5mg/ml、5mg/ml、10mg/ml、15mg/ml、20mg/ml)时的OD值较未加PMMA颗粒的对照组均有增高。PMMA对细胞ALP比活性的影响呈双向改变,当PMMA浓度为0~25μg/ml时,ALP比活性随着PMMA浓度的升高而升高,当PMMA浓度为25~200μg/ml时,ALP比活性随着PMMA浓度的升高而降低,且呈浓度依赖性。结论:离体条件下PMMA对MC3T3-E1细胞的增殖有促进作用,对ALP活性具有双向影响。Aims:To study the effect of polymethyl methacrylate(PMMA) particles on the proliferation,differentiation and function of MC3T3-E1 osteoblast(OB) cell lines.Methods:Cell proliferation was detected with MTT assay.Alkaline phosphatase(ALP) activity of MC3T3-E1 cells was detected with kit.Result:When PMMA particle concentrations were 2.5 mg/ml、5 mg/ml、10 mg/ml、15 mg/ml、20 mg/ml,OD values were higher than that of the control group.PMMA had bipartite effect on the ALP activity,when PMMA concentration was 0~25?g/ml,ALP activity increased in according with the PMMA concentration,when PMMA concentration was 25—200?g/ml,ALP activity decreased in according with the PMMA concentration in a dose-depend manner.Conclusion:In vitro study,PMMA could significantly increase the cell proliferation while it had bipartite effect on the ALP activity in cultured MC3T3-E1 cell line.
