摘 要:目的:探讨大剂量通冠胶囊对动脉粥样硬化兔血一氧化氮(NO)、假血友病因子(vWF)水平及血管内膜、中膜厚度的影响。方法:健康雄性新西兰大耳白兔18只,随机分为空白对照组、高脂组、药物组(通冠胶囊组)共3组各6只,通过免疫性内皮损伤合并高脂饲料诱导法建立动脉粥样硬化兔实验动物模型,对照组喂饲普通饲料,高脂组喂饲高脂饲料,药物组在高脂组的基础上每天加喂通冠胶囊15mg/kg,于实验6周末测定血中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、NO、vWF,结束后取左髂外动脉约2cm行血管内皮病理学检测,计算血管腔内膜和中膜的厚度。结果:药物组TC、TG、LDL-C、vWF水平及内膜厚度均低于高脂组(P<0.05);中膜厚度低于对照组及高脂组;HDL-C、NO水平均高于对照组及高脂组(P<0.05)。结论:通冠胶囊能够降低内皮损伤后血vWF水平,同时提高血NO水平,减少血管内膜、中膜厚度,从而抑制内膜增生及血管狭窄。Objective: To study the effect of Tongguan capsule on serum nitric oxide (NO) and von Willebrand factor (vWF) levels, and arterial intima and media thickness in rabbits with atherosclerosis. Methods.. Eighteen healthy male New Zealand rabbits were randomly divided into three groups, including blank control group, high-fat group, and drug group (Tongguan capsule group). Atherosclerotic rabbit model was established by immune-induced endothelial injury and by feeding with high-fat diet. The control group was given ordinary forage, the high-fat group was given high-fat food based on ordinary forage, and the drug group was given 15mg of Tongguan capsule daily on the basis of food in-take like high-fat group. Serum.total cholesterol (TC), triglyceride(TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), NO and vWF levels were detected on the sixth week of experiment. At the end of experiment, a 2-cm external lilac artery segment was taken out for the endothelial pathological examination, and the arterial intima and media thickness of external lilac artery was calculated. Results: Serum TC, TG, LDL-C, and vWF levels as well as the arterial intima thickness of the drug group were significantly lower than those of the high-fat group, the arterial media thickness was lower while HDL-C and NO levels were significantly higher than those of the control group and the high-fat group. Conclusion-Tongguan capsule can inhibit hyperplasia of the arterial intima and vascular stenosis by reducing the vWF level of the injured endothelia, increasing the NO level, and decreasing the arterial intima and media thickness.
