摘  要:目的观察青蒿提取物对房颤大鼠心肌炎症相关因子肿瘤坏死因子-α(TNF-α)及细胞间黏附分子-1(ICAM-1)表达的影响,探讨青蒿抗心律失常的作用机制。方法将大鼠分为对照组、模型组、青蒿组、卡托普利组。采用氯化钙、乙酰胆碱混合液以舌下静脉给药的方式建立房颤动物模型,隔日重复1次,连续4周,停药后心电图仍有典型房颤改变为造模成功。实验当日,动物静脉血,ELISA法测样品中TNF-α、ICAM-1浓度。结果模型组血清TNF-α、ICAM-1水平较对照组均有提高(P<0.05)。卡托普利组在降低TNF-α、ICAM-1表达方面作用最为显著,青蒿组作用略差于卡托普利组(P>0.05)。结论青蒿提取物通过抑制炎症因子TNF-α、ICAM-1,抑制心脏结构重构和电重构,减少心律失常的发生。Objective To observe the effect of artemisia annua L.extract(AALE) on expression of tumor necrosis factor-α(TNF-α) and intercellular adhesion molecule-1(ICAM-1) in rats with atrial fibrillation,and to explore anti-arrhythmic mechanism of artemisia annua.Methods Sixty Sprague-Dawley(SD) rats are divided into the control group(n=15),the model group(n=15),AALE group(n=15),and captopril group(n=15).Models with atrial fibrillation were established by sublingual intravenous administration of calcium chloride and acetylcholine mixture.The concentrations of serum TNF-α and ICAM-1 were tested by ELISA.Results The levels of serum TNF-α and ICAM-1 in model group were significantly increased than that in the control group(P0.05).The expression of TNF-α and ICAM-1 in captopril group was most significant,and that in AALE group was slightly lower than that in captopril group.There was no significant difference between these two groups(P0.05).Conclusion Artemisia annua L.extract might inhibit cardiac structural remodeling and electrical remodeling,and reduce the incidence of arrhythmias by inhibiting the inflammatory cytokines TNF-α,ICAM-1.
