摘  要:目的构建一种操作简便、重复性高的小动物带瓣管道异种异位移植模型。方法实验组采用四分枝聚乙二醇交联去细胞的sD大鼠肺动脉带瓣管道作为移植物,以端端吻合方式间断缝合于新西兰白兔右颈总动脉,术后第7、14、28天行多普勒超声检查管道通畅情况,28d后取出管道行石蜡切片形态学检查。对照组大鼠接受假手术。结果手术成功率实验组96.0%(24/25),对照组100.O%(20/20),28d实验组移植管道通畅率87.5%(21/24),对照组吻合VI通畅率95.0%(19/20),两组间差异均无统计学意义(P〉0.05),苏木素-伊红(HE)染色可见局部无明显炎症细胞浸润,瓣膜形态、结构完整。结论此模型操作简便、构建迅速、成功率高,有助于检测组织工程瓣膜在体性能。Objective To explore a simple and repeatable method for establishing an in vivo animal model of tissue-engineered valved conduit. Methods Forty-five adult female New Zealand white rabbits, weighing 2. 5-3.0 kg, were assigned randomly into two groups : experimental group ( n = 25 ) and control group (n = 20). Deeellularized valved conduits harvested form female SD rats' aorta were crosslinked with branched poly (ethylene glycol) and then were end-to-end anstomosised with interrupted suture into the right common carotid artery of rabbits in the experimental group. Sham operations were performed in the control group. The patency rate of the transplants was tested in vivo by color Doppler ultrasound on the postoperative day 7, 14 and 28. Then the transplants were removed to be analyzed by hematoxylin and eosin (HE) staining. Results There was no significant difference in the success rate between the experiment group and control group (96.0% vs. 100.0% ,P〉0.05) as well as the patency rate (87.5% vs. 95.0% ,P 〉0. 05). HE staining showed little presence of inflammatory cells and structural integrity of the valves. Conclusion The results suggest that this novel model could provide a convenient and economical way with high success rate for estimating the in vivo functions and characteristics of tissue engineering valved conduit.
