摘 要:采用AFLP技术分别对太湖、长江日本沼虾(Macrobrachium nipponense)群体各30尾的基因组DNA进行了群体特异性位点比对分析,并对各位点基因频率分布进行正态性检验、方差齐性检验(F检验)、样本平均数差异显著性检验(t检验)。结果显示:3对引物共扩增出138个位点;太湖组是独立于长江组的群体,而且它们是两个分化了的野生群体;采用变异性的显著性检验(χ2检验)对位点进行差异比较发现,23个位点在太湖组中出现的频率显著高于长江组,其中位点BY160、BY320、BY470、BY520是与太湖群体分化显著关联的位点,可作为区别太湖野生群体与长江野生群体的标记。In this study,the amplified fragment length polymorphism(AFLP) was used to analyze the specific loci of the two populations(30 samples for each) of Macrobrachium nipponense from the Taihu Lake and the Yangtze River.A total of 138 loci were amplified with 3 pairs of primers.Normality test,homogeneity of variance test(F test) and significant difference between two samples mean test(t test) of loci frequencies were conducted.The statistical results showed that these two populations were different from each other,and they were two differentiated wild populations.χ2 test was used to detect the differences between sites.The results showed that the frequencies of 23 loci were significantly higher in Taihu lake population than that in Yangtze river population(P0.01),especially for loci BY160,BY320,BY470,BY520,which were population-specific loci for the Taihu Lake population.These three loci can be used as specific markers to identify wild populations between from the Taihu Lake and the Yangtze River.
