摘  要:从患内脏白点病的斑鳢Channa maculata肝、脾、肾组织中分离并筛选出一株致病菌GC1,对该病菌的形态学及生理生化特性进行了测定,结果均符合舒氏气单胞菌Aeromonas schubertii的特性。以细菌16S rRNA基因通用引物进行PCR扩增,得到该菌的部分16S rRNA基因序列,长约1 502 bp。将所测序列与GenBank中序列进行Blast比对并构建系统进化树,结果表明,该序列与舒氏气单胞菌的同源性高达99%。人工感染试验证明,该菌株具有极强的致病力,LD50为104.5CFU/mL。29种药物筛选结果显示:该菌对菌必治、先锋必、头孢氨苄、头孢噻吩、先锋V、萘啶酸、四环素、氧氟沙星、氟罗沙星等18种药物高度敏感;对阿莫西林、红霉素、克拉霉素等7种药物中度敏感;对青霉素G、杆菌肽、苯唑青霉素等不敏感。A pathogenic bacterial strain GC1 was isolated from the liver,spleen and kidney of the diseased Taiwan snakehead Channa maculata,and identified as Aeromonas schubertii according to its morphologic and physio-biochemical characteristics.Using the general primers of bacterial 16S rRNA,the gene fragments were amplified by PCR with about 1502 bp,and the obtained gene sequence was compared with the one in GenBank on the base of 16S rRNA.The Blast showed that GC1 had the maximum homologies(i.e.,99%) with the sequence of A.schubertii and the phylogenetic tree further revealed that this strain clustered together with A.schubertii.Furthermore,the artificial infection tests verified that GC1 had a strong virulence,with LD50 of 104.5CFU/mL.By testing with 29 medicaments,high sensitivity was found to Cet riaxone,Cefobid,Cefalexin Granules,Cefalotin,Cefalin V,Nalidixic acid,Tetracycline,Ofloxacin and Fleroxacin;however,the strain GC1 was shown to be insensitive to Penicillin G,bacitracin and Oxacillin.
