摘  要:目的研究磷脂类代谢产物1-磷酸鞘氨醇(S1P)对血小板活化的影响。方法利用酶联免疫法检测血小板中S1P的含量,利用流式细胞仪检测血小板凋亡、活化状况。结果随着保存时间的延长,血小板悬液中的S1P含量(μmol/L)持续增加,从第1天至第5天分别为:1.38±0.17、2.07±0.25、2.78±0.31、3.53±0.28、4.56±0.46。加入不同浓度的外源性S1P,对保存5d期间的血小板生理指标进行动态监测。不同处理组间,血小板的活化率随加入的S1P浓度的增加而升高,凋亡程度增加,血小板结构发生改变。上述实验结果的差异均有统计学意义(P<0.01)。结论随着血小板保存时间的延长,血小板血浆中S1P含量逐渐增加,血小板功能逐渐降低。提示血小板释放的有生物活性的磷脂代谢产物可能对血小板的功能施加某种影响,可能是引起血小板输注无效的一个重要原因。Objective To study the activated platelet delivers the sphingosine 1 phosphate,which has some influence on platelet′s function during conservation.Methods Using enzyme linked immunosorbent assay to observe the create regularity of S1P.Activation and apoptosis of platelet were detected by flow cytometry.Results As to conservation time,the content of S1P continued to increase.The contents of S1P(μmol/L)were 1.38±0.17,2.07±0.25,2.78±0.31,3.53±0.28 and 4.56±0.46 from 1 d to 5 d preservation respectively.We observed the changes of platelet′s function after adding exogenous S1P with different concentration too.The results displayed that the ratio of activation and apoptosis increased with S1P concentration,and had disparity in different groups.Platelets′ structure changed too.Conclution As extended to save time,S1P level in platelets′ plasma gradually increase,and platelets′ function gradually reduce.The results suggest that phospholipid metabolism with biological activity released by platelet may exert a certain influence on platelet function,which could be an important reason to cause platelet transfusion refractoriness.
