摘 要:目的研究溃结2号方对实验性溃疡性结肠炎(UC)大鼠结肠黏膜组织肿瘤坏死因子-α(TNF-α)及其mRNA表达的影响。方法采用2,4,6-三硝基苯磺酸(2,4,6-TNBS)+免疫诱导法复制UC大鼠模型,随机分组为模型组、中药组、SASP组,分别给予相应的干预。采用免疫组织化学PAP二步法检测大鼠结肠组织中TNF-α的表达,采用实时荧光定量法(RT-PCR)检测TNF-αmRNA的水平,并观察大鼠治疗14天及21天后TNF-α及mRNA的表达情况。结果连续灌胃14天后,与模型组比较,中药组、SASP组大鼠结肠黏膜TNF-α水平降低,差异有统计学意义(P<0.05);中药组与SASP组TNF-α水平差异无统计学意义(P>0.05);各组之间TNF-αmRNA水平差异无统计学意义(P>0.05)。连续灌胃21天后,与模型组比较,中药组、SASP组大鼠结肠黏膜TNF-α水平、TNF-αmRNA水平差异有统计学意义(P<0.01,P<0.05);与SASP组比较,中药组TNF-α含量降低,差异有统计学意义(P<0.05)。中药组灌胃21天后,TNF-α水平、TNF-αmRNA水平明显低于灌胃14天,差异有统计学意义(P<0.05)。结论溃结2号方不仅能下调UC大鼠结肠黏膜TNF-α的水平,还能抑制TNF-αmRNA的表达,说明溃结2号方治疗UC的作用机制和下调作用TNF-α密切相关。Objective To study the effect of "No.2 Kuijie Decoction" on expression of TNF-α and TNF-α mRNA in ulcerative colitis(UC) rats.Methods The rats were duplicated by 2,4,6-trinitrobenzene sμlfonic acid(TNBS) and immune induced methods,then randomized into model group,Chinese medicine group and SASP group.The expression of TNF-α was detected by immunohistochemistry method and the level of TNF-α mRNA was detected by RT-PCR method.The expressions of TNF-α and TNF-α mRNA were observed on 14th day and after treatment of 21 days.Results After oral irrigation for 14 days,the TNF-α was lower in Chinese medicine group and SASP group than the model group,with significant difference(P0.05);there was no significant difference in TNF-α level between Chinese medicine group and SASP group(P0.05);there were no significant differences in TNF-α mRNA level among the groups(P0.05).After oral irrigation for 21 days,the levels of TNF-α and TNF-α mRNA in Chinese medicine group and SASP group were significantly different from the model group(P0.01,P0.05);the TNF-α was lower in Chinese medicine group than that in SASP group,with statistical difference(P0.05);the levels of TNF-α and TNF-α mRNA were lower on 21 days than 14 days,with significant difference(P0.05).Conclusion "No.2 Kuijie Decoction" can low the TNF-α level and restrain the expression of TNF-α mRNA in UC rats.The mechanism of "No.2 Kuijie Decoction" on UC rats is associated with TNF-α.
