摘　　要：目的:研究骨桥蛋白(osteopontin,OPN)在原发性干燥综合征(primary Sjgren’s syndrome,pSS)患者唇腺组织中的表达情况。方法:收集24例pSS患者和14例对照者的唇腺标本,提取唇腺中总RNA,利用反转录聚合酶链反应(RT-PCR)法扩增OPN、β-肌动蛋白(β-actin)的片段,计算OPN/β-actin光密度比值。采用免疫组织化学染色方法检测OPN蛋白在唇腺中的表达情况。结果:pSS组唇腺中OPN的mRNA转录及蛋白表达水平均高于对照组(P<0.05)。OPN表达水平与淋巴细胞灶性评分(lymphocyte foci-score,LFS)呈正相关,mRNA转录水平与LFS相关系数为0.407(Pearson相关系数),P=0.049;蛋白表达程度与唇腺LFS相关系数为0.476(Spearman相关系数),P=0.039。免疫组织化学染色显示,OPN蛋白主要分布在腺上皮细胞胞浆内,但在浸润的淋巴细胞胞浆中也有阳性表达。结论:pSS患者唇腺中OPN表达增加,可能是唇腺结构破坏的原因之一。Objective：To investigate the mRNA transcription and protein expression of osteopontin（OPN） and to analyze the possible role of OPN in primary Sjgren＇s syndrome（pSS）.Methods： In this study,24 patients with pSS were selected and diagnosed according to the American-European Consensus Group criteria.The control group was composed of 14 subjects.Total RNA from labial glands was extracted.The target gene and β-actin acted as templates to perform reverse transcript polymerase chain reaction（RT-PCR）.Then mRNA expression of the target gene were semi-quantitated by the OD ratio of the target gene to β-actin bands on gel under densitometry.Immunohistocheical analysis was used to determine the expression of proteins of the target genes in labial glands from patients and healthy control.Results： In contrast to the control group,the mRNA transcription and protein level of the target genes in labial glands in pSS patients were significantly increased statistically（P0.05）.Spearman and Pearson rank correlation coefficients were used for analyzing the correlation of lymphocytes foci-score（LFS） with OPN expressions,the level of the mRNA transcription（0.407,P=0.049） and protein expression of OPN（0.476,P=0.039） was positively correlated with the LFS.Immunohistochemistry detected that OPN protein was not only mainly expressed in intracytoplasm of the gland duct,but also in infiltrated lymphocytes.Conclusion： OPN is higher expressed in the basal and surface of labial glands of pSS patients,which may contribute to the destruction of labial glands.
