摘  要:目的:研究尖吻蝮蛇毒抑瘤组分Ⅰ(AAVC-Ⅰ)对小鼠肺癌LLC细胞增殖抑制的影响。方法:以小鼠肺癌LLC细胞为研究对象,采用显微镜观察细胞形态学的变化、MTT法检测药物敏感性、DNA ladder法检测细胞凋亡。结果:AAVC-Ⅰ对LLC细胞有较强的增殖抑制作用,抑制程度随药物浓度的增加而增强,其IC50为43.823μg/ml(P<0.05)。琼脂糖电泳观察到细胞DNA的片段化。结论:AAVC-Ⅰ可抑制LLC细胞增殖,诱导其凋亡。Objective:To investigate the effects of anti-tumor component Ⅰ from Agkistrodon acutus venom(AAVC-Ⅰ) on the cell proliferation and apoptosis of mouse Lewis lung cancer(LLC).Methods:Cell line of LLC was obtained and observed for the morphological change with invert microscope and detected by thiazolyl blue tetrazolium bromide(MTT) for the cell sensitivity to AAVC-Ⅰ.DNA ladder method was used to examine the cell apoptosis.Results:AAVC-Ⅰ produced strong inhibition on LLC cells in vitro,and the cyto-inhibition was enhanced with drug concentration,with a IC50(50% concentration of inhibition) of 43.823 μg/ml(P0.05).In addition,DNA fragments were seen using agarose gel electrophoresis.Conclusion:AAVC-Ⅰ can inhibit LLC value and facilitate apoptosis of the cell line.
