摘 要:血虚症是祖国医学体系重要的征候群之一,其发病机制尤其是分子机制的阐明对于深入认识血虚症及相关疾病具有重要意义.细胞凋亡介入了多种生理、病理过程,与造血器官的发育、成熟及血液系统病密切相关.本研究旨在探讨造血细胞凋亡与血虚症的关系.选择成年C57B/6J小鼠作为实验对象,经不同剂量(2 .5、5. 5、7.5Gy)γ射线照射后,小鼠在 7d左右表现出不同程度的血虚症:各剂量组外周血白细胞均明显下降,小鼠皮毛干燥,光泽差,懒动.形态学检查显示,照后各剂量组小鼠的骨髓和脾脏细胞出现典型的凋亡变化,如核固缩、核碎裂及形成凋亡小体等,这些变化以照后 24h之内最明显,随后凋亡细胞逐渐减少, 7d左右基本消失;相应地,照后24h之内各剂量组小鼠骨髓和脾脏细胞DNA凝胶电泳出现明显的梯状带,随着时间的推移梯状带减弱、消失.上述造血细胞形态学及DNA的变化与血虚症的严重程度呈正相关,即细胞凋亡越明显,小鼠血虚症越严重,但造血细胞凋亡出现的时间与血虚症的程度无关,即无论照射剂量大小,凋亡都发生在血虚症出现的早期.结论 造血细胞凋亡与血虚症的发生、发展密切相关,可能是始动环节之一.In order to investigate the involvement of apoptosis of hemopoietic cells during the process of blood deficiency(BD), a BD model of mouse with different levels of seventy was produced by radiation with different doses(2. 5Gy .5. 5Gy and 7. 5Gy) of γ- ray. Then, the apoptotic cells from bone marrow and spleen of mice were identified in morphology with microscope and in biochemistry with DNA gel elctrophoresis. Seven days after radiation, the indications of BD including decrease of peripheral white blood cells, indolence in action and wizened fur appeared significantly in mice. Within 24 hours after radiation, a large amount of apoptotic cells existed in tissues of bone marrow and spleen, afterwards the apoptotic cells gradually decreased. Occurring time of these changes was does-independent,otherwise, the numbers of apoptotic cells were dependent on the seventy of BD. In conclusion, apoptosis of hemopoietic cells is early involved in the process of BD and likely plays a key role during the process.
