摘 要:目的采用病例对照研究探讨中国汉族人群磷酸二酯酶4D(phosphodiesterase 4D,PDE4D)基因与动脉粥样硬化血栓形成性脑卒中(atherothrombosis ischemic stroke,AIS)的关系。方法应用聚合酶链反应—限制性片段长度多态性方法,检测235例浙南地区汉族人群AIS患者和105例非脑卒中对照组PDE4D基因SNP83、32酶切多态性。结果 AIS患者PDE4D基因SNP83 C等位基因频率明显高于对照组,是AIS的危险因素(OR=1.718,95%CI:1.120~2.633;P=0.012);CC+CT基因型亦明显高于对照组,差异有统计学意义(P<0.05),(OR=1.875,95%CI=1.137~3.092);调整糖尿病、HDL-C等危险因素后,差异仍存在(OR=2.023,95%CI:1.207~3.391)。PDE4D基因SNP32位点在两组比较无统计学意义(P>0.05)。结论对于中国汉族人群,PDE4D基因SNP83是AIS的危险因素,而SNP32与AIS无关联。Objective To investigate the association between 2 single nucleotide polymorphisms(SNPs) of the phosphodiesterase 4D (PDE4D) gene and atherothrombosis ischemic stroke (AIS)in the Hau population in China. Methods Two polymorphisms SNP83 and SNP 32 in PDE4D gene were analyzed in 235 patients of AIS and 105 non - cerebral infarction controls from south of Zhejiang Province using polymerase chain reaction - restriction fragment length polymorphism method in a case - control study. Results The C allele frequency of SNP83 in cases was higher than controls. The C allele was a risk factor for AIS ( OR = 1. 718,95% C1 : 1. 120 - 2. 633 ; P = Genotype frequencies between cases and controls at SNP83 showed significant differences. The( CC + CT) genotypes had an OR of P = 0. 008) for cases compared with the TT genotype. No association of the SNP32 in PDE4D with A1S was observed in our study. on SNP83 in the PDE4D gene is associated with AIS in the Chinese Hart population.
