摘 要:目的:优选淫羊藿总黄酮的提取纯化工艺。方法:采用L9(34)正交设计法,以浸膏得率和总黄酮含量为检测指标优选提取工艺;用AB-8型大孔吸附树脂柱进行分离,乙酸乙酯、正丁醇萃取纯化。结果:淫羊藿总黄酮的最佳提取工艺为淫羊藿叶的粗粉中加入27倍量水(V/W),浸泡2 h后,煎煮3次,每次1 h;淫羊藿提取液上AB-8型大孔吸附树脂柱分离,再用乙酸乙酯、正丁醇萃取,所得淫羊藿总黄酮的平均含量(以淫羊藿苷计)为60.91%。结论:该工艺稳定可行,提取淫羊藿总黄酮的含量较高,可为工业生产提供依据。Objective: To optimize extraction and purification process of total flavonoids from Herba Epimedii.Methods: The optimum extractive technology was investigated by using L9(34) design with the extracting rate and the contents of total flavonoids as evaluation indexes.AB-8 macroporous resign column was used to isolate flavonoids,and ethyl acetate,N-butanol was used to extract and purify flavonoids.Results: The optimum extraction condition was: adding 27 times amount of water to Herba Epimedii leaf powder,soaking 2h,boiling 3 times each time for 1h,loading epimedium extracts into AB-8 macroporous resign column,and separating with ethyl acetate and N-butanol.The average content of total flavonoids from epimedium(in terms of icariin) was 60.91%.Conclusion: This process is stable,feasible,and the extraction of epimedium flavonoids contents is higher,which may provide reference for industrial production.
