摘 要:目的研究异叶败酱提取物作用于人白血病K562细胞的分子机制。方法以异叶败酱提取物作用于K562细胞后提取细胞总蛋白,用双向电泳对蛋白进行分离,对异叶败酱提取物处理前后的K562细胞总蛋白双向电泳图谱进行差异分析,通过质谱技术鉴定差异明显的蛋白质。结果通过对比分析异叶败酱提取物处理前后的白血病K562细胞总蛋白双向电泳图谱,发现异叶败酱提取物处理组中有23个蛋白点发生了明显改变。经质谱和Mascot软件分析,成功鉴定了3个蛋白质,分别为热休克蛋白90β,甘油醛-3-磷酸脱氢酶,血红蛋白。结论通过蛋白质组学技术,发现了异叶败酱提取物处理前后白血病K562细胞差异明显的蛋白质点23个,这些差异蛋白质可能是异叶败酱提取物对白血病K562细胞产生增殖抑制、凋亡诱导和诱导分化作用的关键蛋白质。Objective To study the molecular mechanism of the extracts of Yiyebaijiang(Patrinia heterophylla) acting on human leukemia K562 cells.Methods The total protein of K562 cells was extracted after K562 cells were acted by the extracts of Yiyebaijiang.The proteins were separated by using two-dimensional electrophoretogram(2-DE).The electrophoretogram of the total protein of K562 cells were given variance analysis before and after Yiyebaijiang treatment.The proteins with obvious differences were identified by using mass-spectrometric technique.Results There were 23 protein points having significant changes in the group treated with the extracts of Yiyebaijiang(trial group) through comparing and analyzing 2-DE of the total protein of K562 cells before and after Yiyebaijiang treatment.After the analyses of mass-spectrometric technique and Mascot software,there were 3 proteins identified successfully,including heat shock protein 90β,glyceraldehyde-3-phosphate dehydrogenase and hemoglobin.Conclusion There have been 23 protein points with obvious differences found out in K562 cells before and after Yiyebaijiang treatment through proteomics technique.These differential proteins may be key ones for the effects of the extracts of Yiyebaijiang in inhibiting proliferation and inducing apoptosis and differentiation in K562 cells.
