摘  要:目的鉴定和特征描述海南岛蚊子基因组弹状病毒核蛋白内生病毒片段。方法 收集海南岛内的库蚊、伊蚊和按蚊,分别提取各类蚊子的总基因组,以此为模板PCR扩增弹状病毒类核蛋白内生病毒片段,并通过测序和分子生物学方法对其特征进行鉴定和描述。结果 在海南岛库蚊和伊蚊基因组中获得弹状病核蛋白内生病毒片段,采用测序和分子生物学方法分析和鉴定了该内生病毒片段在进化上高度保守。结论 数据分析证明蚊子基因组弹状病毒核蛋白内生病毒片段与弗兰德斯病毒(Flanders virus)相关,这种病毒在海南岛还没见报导过;初步表明蚊子基因组弹状病毒核蛋白内生病毒片段是在蚊子扩散到海南岛来之前就已经整合到蚊子基因组当中的。Objective To identify and characterize rhabdovirus nucleoprotein (NP)-like endogenous viral elements (EVEs) in the genomes of mosquitoes from Hainan Island.Methods Wild mosquito samples were collected from Hainan Island and their total DNA was isolated from these moquitoes as a template for amplifying Rhabdovirus NP-like EvEs,which were identified and characterized by PCR methods and bioinformatic analysis.Results Rhabdovirus NP-like EVEs amplified by PCR showed positive in the genomes of Aedes aegypti and Culex quinquefasciatus mosquitoes but negative in Anopheles sinensis mosquitoes.Sequencing analysis revealed that the amplified sequence was homologous to the NP gene of Flanders virus.Rhabdovirus NP-like EVEs contained an intact open reading frame (ORF) and were expressed as mRNA.Phylogenetic analysis showed that rhabdovirus NP-like EVEs derived from mosquitoes grouped into a clade and were closely related to Rabies virus and Drosophila sigma virus.Conclusions Rhabdovirus NP-like EVEs are found to be related to Flanders virus,which has never been isolated in Hainan Island.NP-like EVEs might be integrated into the mosquito genomes prior to its dispersal.
