乳安凝胶膏剂对大鼠乳腺增生及内分泌激素的影响

摘　　要：目的观察乳安凝胶膏剂对大鼠乳腺增生的作用并探讨其作用机制。方法 56只雌性未孕大鼠随机分为7组,正常对照组、乳腺增生模型组、赋形剂对照组、阳性对照组、乳安凝胶膏剂4.16、2.08、1.04 g.kg-1剂量组,采用注射苯甲酸雌二醇、黄体酮造模,给药30 d。观察大鼠乳头直径,血清孕酮(P)、雌二醇(E2)、泌乳素(PRL)水平,乳腺组织病理形态。乳安凝胶膏剂4.16、2.08 g·kg-1组能降低模型大鼠的乳头直径,4.16 g·kg-1组能降低血清E2、PRL水平,提高血清P水平,缩小模型鼠乳腺小叶体积、腺泡腔,减少乳腺腺泡数量、分泌物,与模型组比较,差异均有统计学意义(P<0.05,0.01)。结论乳安凝胶膏剂可能通过调节乳腺增生大鼠的性激素水平,改善增生乳腺的病理形态学变化而对抗雌激素引起的大鼠乳腺增生。Objective To observe the Ru-an gel-unguentum effect on hyperplasia of mammary glands with rats and to investigate the mechanism of Ru-an gel-unguentum.Methods Fifty-six female unpregnancy rats were divided into 7 groups at random：normal control group,cyclomastopathy model group,negative control group,positive control group,group with high dose of Ru-an gel unguentum,group with medium dose and group with low dose.And rats were injected estradiol benzoate and progesterone to be made inyo cyclomastopathy models,and the administration time was 30 days.Detailed observation was carried out on the level of progesterone,estradiol and prolactin in blood serum of rats,the papilla diameter of rats and the pathology modality of breast tissues.Results The papilla diameter of rats would be decreased with high and medium dose of Ru-an gel-unguentum.With high dose of Ru-an gel-unguentum,the levels of estradiol and prolactin were dereased,the level of progesterone was increased and the volumes of mammary gland lobules and acinar lumina in models were decreased,the quantity of mammary glands acinus and secretions were decreased.Compared with the cyclomastopathy model group,the difference is statistically significant.Conclusion Ru-an gel-unguentum accommodates sex hormone levels of rats cyclomastopathy,and ameliorates the pathology modality of breast tissues,so as to be against rat cyclomastopathy caused by estrogen.
