HPLC法同时测定宣木瓜中绿原酸、芦丁、金丝桃苷、槲皮素的含量

摘　　要：目的建立宣木瓜药材中绿原酸、芦丁、金丝桃苷、槲皮素的含量测定方法。方法采用HPLC法,Phenomenex luna C18色谱柱(250 mm×4.6 mm,5μm),流动相为0.6%磷酸溶液(A)和乙腈(B),梯度洗脱,检测波长为360 nm,流速设为1.0 ml.min-1。结果绿原酸、芦丁、金丝桃苷、槲皮素线性范围分别为0.035～0.56 g·L-1(r=0.997 8),0.003 2～0.051 2 g·L-1(r=0.999 3),0.004 25～0.068 g·L-1(r=0.999 7),0.003 5～0.056 g·L-1(r=0.999 7),平均加样回收率分别为102.9%(RSD=1.04%)、99.3%(RSD=2.28%)和99.2%(RSD=2.33%)、99.5%(RSD=2.52%)。结论该方法操作简单,结果准确,具有较好的重复性和稳定性,可用于宣木瓜药材的质量控制。Objective To establish an HPLC method for the simultaneous determination of chlorogenic acid,rutin,hyperoside and quercetin in Fructus Chaenomelis.Methods Ahigh-performance liquid chromatography equipped a Phenomenex luna C18（250 mm×4.6 mm,5 μm） column with UV detection used.The mobile phase consisited of 0.6% aqueous phosphoric acid（A） and acetonitrile（B） with gradient elution.The detection wavelength was set at 360 nm and the flow rate was 1.0 ml·min-1.Results The linear ranges of chlorogenic acid,rutin,hyperoside and quercetin were 0.035~0.56 g·L-1（r=0.997 8）,0.003 2~0.051 2 g·L-1（r=0.999 3）,0.004 25~0.068 g·L-1（r=0.999 7）,0.003 5~0.056 g·L-1（r=0.999 7）,respectively;the average recoveries were 102.9%（RSD=1.04%）,99.3%（RSD=2.28%）,99.2%（RSD=2.33%）,99.5%（RSD=2.52%）.Conclusion The proposed method is simple,accurate,and repeatable.which can be used for quality control of Fructus Chaenomelis.
