摘  要:目的探讨不同剂量的十一酸睾酮对生精细胞凋亡的影响,寻找更好的节育方法。方法将雄性小鼠随机分为4组,低、中、高剂量给药组3组和对照组,给药组每2周1次在大腿内侧肌肉注射十一酸睾酮,低、中、高剂量组每次注射剂量分别为0.25、0.5、0.75 mg/kg;对照组注射安慰剂(即精制茶油0.5mg/kg),共3个月,末次给药2周后,内眦静脉取血,检测血睾酮的含量;用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL)检测睾丸生精细胞凋亡。结果实验组各组血睾酮差异无统计学意义,与对照组比血睾酮差异也无统计学意义;实验组的细胞凋亡率显著高于对照组(P<0.05)。结论过量摄入十一酸睾酮引起雄性小鼠生精细胞凋亡增加,临床可考虑据此找到更好的节育办法。Objective To explore the different doses of testosterone undecanoate on spermatogenic cell apoptosis in rats and search for a better method of birth control.Methods Male mice were randomly divided into four groups,low,medium,high dosage group and the control group,every two weeks testosterone undecanoate were given in the medial thigh muscle in low,medium,high dose group,the injection doses were respectively 0.25,0.5,0.75 mg / kg;the control group was injected with placebo(i.e.,refined tea oil 0.5mg / kg),a total of three months,2 weeks after the last administration,medial vein blood sampling,detection of blood testosterone levels;with DNA terminal transferase mediated nick end labeling(terminal deoxynucleotidul transferase mediated nick end labeling,TUNEL) detection of testicular spermatogenic cell apoptosis.Results The experimental group blood testosterone had no significant difference compared with that of the control group,the blood testosterone also showed there was no significant difference between the experimental group;the apoptosis rate was significantly higher than that in the control group(P 0.05).conclusion Excessive intake of testosterone undecanoate may induced male mouse spermatogenic cell apoptosis,which may be considered accordingly to find better methods of birth control.
