益气活血方对心肌缺血大鼠心功能及心肌闰盘相关基因表达的影响

摘　　要：目的观察益气活血方对心肌梗死模型大鼠心功能及心肌闰盘相关基因表达的影响。方法 120只SD大鼠随机分为6组,假手术组、模型对照组、阳性对照组及益气活血方低、中、高剂量组,连续7次给药后,结扎冠状动脉复制急性心肌梗死模型,4h后经右颈总动脉行左心室插管,测定左心室压力峰值(LVP)等心功能指标;取心肌组织,PCR法检测闰盘桥粒斑蛋白、N-钙黏蛋白和心肌缝隙连接蛋白(Cx43蛋白)的mRNA表达。结果与模型对照组比较,益气活血方中、高剂量组LVP明显提高(p<0.05),高剂量组+LVdp/dt_(max)、-LVdp/dt_(max)明显升高(p<0.05)。与模型对照组比较,低剂量组桥粒斑蛋白、N-钙黏蛋白、Cx43 mRNA的表达水平改变不明显,中剂量组和高剂量组表达水平明显升高。结论益气活血方对急性心肌缺血大鼠心功能有明显的改善作用,其作用可能与升高心肌闰盘桥粒斑蛋白、N-钙黏蛋白、Cx43 mRNA的表达、改善心肌细胞间信息传递有关。Objective To investigate the effects of Yiqihuoxue(YQHX) decoction on cardiac function and mRNA expression of intercalated disc related protein in rats with myocardial ischemia.Methods 120 Sprague Dawley(SD) rats were randomly divided into six groups;sham operation group,model control group,positive control group,the YQHX decoction groups with low,medium and high dose,which were administered for 7 times and the acute myocardial infarction model was developed with coronary artery ligation.After 4h for the surgery,left ventricular peak pressure(LVP) and other cardiac function indexes were detected through right common carotid artery cannulation to the left ventricular.The cardiac muscle was extracted to observe the mRNA expression of desmoplakin,N-cadherin,connexin 43(Cx43) with PCR.Results Compared with the model control group,the middle and high dose of the YQHX decoction could improve the LVP(P<0.05 ),and the indexes of + LVdp/dt_(max),-LVdp/dt_(max) were obviously increased in high dose of the YQHX decoction group(P <0.05 ).Comparison to model control group,the expression of desmoplakin,N-cadherin,Cx43 were increased significantly in middle and high dose of YQHX decoction,while no significant change in low dose of YQHX decoction group.Conclusion The YQHX decoction could improve cardiac function of the rats suffered with acute myocardial ischemia,and the mechanism may be related with increasing the expression of desmoplakin,N-cadherin,Cx43 of myocardial cells of intercalated disk.
