摘  要:目的探讨经鼻给予神经生长因子(nerve growth factor,NGF)对实验性创伤性脑外伤(tramatic brain injury,TBI)大鼠大脑的保护作用。方法将大鼠随机分为假手术组、对照组和NGF组,参照Feeney’s自由落体法制作TBI大鼠模型,NGF组经鼻给予NGF治疗。采用平衡木方法评估3组TBI大鼠的神经功能恢复情况。行免疫组化染色检测各组TBI大鼠脑组织β-淀粉样蛋白前体蛋白(β-APP)和神经元特异性烯醇化酶(NSE)的表达情况。结果 NGF组TBI大鼠与对照组相比,运动协调功能恢复较快(P<0.05),免疫组化观察发现对照组和NGF组NSE阳性细胞均明显低于假手术组(P<0.01),NGF组NSE阳性细胞明显高于对照组(P<0.01);对照组和NGF组β-APP阳性表达均明显高于假手术组(P<0.01),NGF组β-APP阳性表达明显低于对照组(P<0.05)。结论经鼻给予NGF可增加TBI大鼠NSE阳性细胞表达,减少β-APP阳性细胞表达,减轻脑水肿,促进损伤后神经功能的恢复。Objective To investigate the protection effect of intranasal nerve growth factor on brain of rats with tramatic brain injury.Methods The rats were randomly divided into sham,control and treatment group.They were subjected to the modified Feeney's weight-drop model.The treatment group was treated with NGF administered by nasal route,and the control group was given phosphate-buffered saline(PBS).Beam walking test was performed in the three groups.Immunohistochemistry was used to detect the β-APP and NSE positive cells near the region of injury in rats after TBI.Results NGF group traversed the beam significantly quicker than control group(P0.05).Immunohistochemical staining showed that NSE was declined significantly in both control and NGF group compared to sham group(P0.01),but it was higher in NGF group compared to control group(P0.01).β-APP was elevated significantly in both control and NGF group compared to sham group(P0.01),but but it was lower in NGF group compared to control group(P0.05).Conclusion Intranasal nerve growth factor after TBI in rats can improve neurol function,increasing NSE positive cells and decreasing β-APP positive cells.
