同种异体大鼠小肠移植中监测CD4^+T细胞和CD8^+T细胞数比值的意义

摘　　要：目的探讨CD4+T细胞数/CD8+T细胞数作为大鼠同种异体小肠移植急性免疫排斥反应指标的可行性。方法选取健康雄性成年SD、Wistar大鼠建立同种异体小肠移植模型,于大鼠术前和术后1、3、5、7、9 d共6个时间段静脉取血,应用免疫荧光染色技术及流式细胞仪检测CD4+T细胞和CD8+T细胞百分率,并计算其比值。同期取移植小肠标本做组织学检测。结果术后CD4+T细胞数和CD8+T细胞数比值前期增高后期降低。组织病理学检测发现术后小肠移植物的排斥反应由轻到重,术后第5天始CD4+T细胞数和CD8+T细胞数比值与急性免疫排斥反应程度呈正相关。结论CD4+T细胞和CD8+T细胞数比值作为大鼠同种异体小肠移植术后急性免疫排斥反应的监测指标的可行性较大。Objective To study the feasibility of the ratio of CD4^＋T cells/CD8^＋T cells as the indicator of acute immune rejection before and after intestinal homografting in rats. Methods The intestinal homografting model was set up in healthy male adult SD and Wistar rats.We checked the positive cell percentage of CD4^＋ and CD8^＋, and its ratio with immunofluoreseence stain- ing technique and flow cytometry before operation and on 1,3,5,7,9 d after operation,as well as the pathologic examination of grafts was conducted to identify the episodes of rejection. The experimental data were analyzed with the statistical software SPSS17.0. Results The ratio of CD4^＋/CD8^＋ after operation was obviously increased in the preceding period and decreased at the later stage. The rejection of intestinal graft was from light to heavy after surgery,through the detection of histopathological tech- niques. Starting from the postoperative 5 d, the ratio of CD4^＋/CD8^＋ was positively correlated with the degree of acute immune re- jection. Conclusion It is of large feasibility that the ratio of CD4^＋/CD8^＋ can be used as an indicator to monitor acute rejection of intestinal homografting in rats.
