急性白血病P16与CDK4蛋白的表达及其临床意义

摘　　要：目的检测急性白血病(acute leukemia,AL)患者外周血单个核细胞中蛋白P16、CDK4缓解前后的表达及临床意义。方法应用免疫组化P-V法,在初治AL组,AL完全缓解(AL-CR)组及健康对照组中,分别检测外周血单个核细胞中P16、CDK4蛋白的表达。结果 P16、CDK4蛋白表达阳性率在健康对照组、AL-CR组及AL组中分别为95.0%,81.2%和41.7%;10.0%,15.6%和64.6%。P16蛋白表达在缓解后增加(P<0.05);CDK4蛋白在缓解后减少(P<0.05)。结论P16、CDK4蛋白表达与AL疗效及预后相关,有助于阐明AL分子生物学机制。Objective To investigate the expression of P16 and CDK4 protein in peripheral blood mononuclear cells of the patient with acute leukemia （AL） before and after remission and its clinical significance. Methods The P-V immunohisto- chemical method was adopted to detect the expression level of P16 and CDK4 protein in peripheral blood mononuclear cells in the AL initial treatment group, AL complete remission （AL-CR） group and the normal control group. Results The positive ex- pression rates of P16 and CDK4 protein were 95.0% and 81.2% in the normal control group,41.7% and 10.0%in the AL-CR group and 15.6% and 64.6% in the AL initial treatment group. Expression of P16 protein was increased after remission （P〈0.05） ; expression of CDK4 protein was decreased after remission （P〈0.05）. Conclusion Expression of P16 and CDK4 protein are relevant with curative effect and prognosis of AL, which will help to clarify the molecular mechanisms of AL.
