摘 要:目的探讨格列吡嗪对健康新西兰白兔心肌缺血预适应的影响。方法以健康新西兰白兔为实验对象,通过开胸结扎左冠状动脉前降支的方法建立心肌缺血预适应的动物模型。将实验动物随机分对照组,预适应组,格列吡嗪组,格列吡嗪+预适应组。实验过程中记录心电图变化,实验结束后,测血肌钙蛋白,取心脏大体TTC染色,计算心肌梗死面积,以肌钙蛋白和心肌梗死面积作为终点效应指标,评价磺脲类药物格列吡嗪对心肌缺血预适应的影响。结果预适应组与对照组相比,实验后血肌钙蛋白、心肌梗死面积较前者明显降低,单用格列吡嗪不影响实验后血肌钙蛋白、心肌梗死面积,格列吡嗪+预适应组与预适应组相比,实验后血肌钙蛋白、心肌梗死面积无明显差异。结论格列吡嗪对健康新西兰白兔心肌缺血预适应没有影响。Objective To investigate the effect of glipizide on myocardial ischemic preconditioning(IPC) in the healthy Newzealand white rabbits. Methods The heathy Newzealand white rabbits as the experimental subjects were performed thoraco- tomy for ligating left anterior descending coronary artery to establish IPC model and then divided into four groups randomly,the control group,preconditioning group,glipizide group and glipizide+preconditioning group. During the experiments, ECG of limb leads were recorded.After the experiments,the sizes of myocardial infarction were calculated by TI'C staining and the concentra- tion of troponin T was detected. The sizes of myocardial infarction and tropnin T were used as the end effect indexes to evaluate the influence of glipizide on IPC. Results The sizes of myocardial infarction and troponin T level were significantly lower in the IPC group than the control group (P〈0.05). Single glipizide did not affect the troponin level and the sizes of myocardial infarction. Com- paring the glipizide+ IPC group with the IPC group,the troponin level and the sizes of myocardial infarction had no significant dif- ferences. Conclusion Glipizide has no effects on IPC in the healthy Newzealand white rabbits.
