摘  要:目的研究幼龄期音乐暴露对成年大鼠焦虑样行为和恐惧记忆消退的影响。方法健康2周龄Sprague—Dawleyda大鼠4窝(共28只),投币法随机分为2组:音乐组每晚8:00—10:00给予音乐暴露,持续3周;对照组不作处理。5周龄后雌、雄鼠分笼,雄鼠音乐组与对照组各8只,雌鼠音乐组与对照组各6只。各组大鼠8周龄时,对其进行恐惧条件化训练、旷场实验、高架十字迷宫实验和恐惧消退训练。结果高架十字迷宫实验结果,电击前音乐组雌、雄大鼠开放臂停留时间百分比[雌鼠(7.07±1.14)%,雄鼠(5.12±1.95)%]与对照组雌、雄大鼠[雌鼠(4.65±0.86)%,雄鼠(4.86±1.95)%]相比差异无统计学意义(P〉0.05);电击后音乐组雌、雄大鼠开放臂停留时间百分比[雌鼠(8.63±3.35)%,雄鼠(7.79±2.49)%]高于对照组雌、雄大鼠[雌鼠(1.48±0.11)%,雄鼠(4.294-1.68)%](尸〈0.01)。旷场实验结果,电击前音乐组雌、雄大鼠中央格停留时间百分比[雌鼠(6.16±2.17)%,雄鼠(6.25±3.47)%]与对照组雌、雄大鼠[雌鼠(5.27±1.95)%,雄鼠(6.22±3.13)%]相比差异无统计学意义(P〉0.05);电击后音乐组雌、雄大鼠中央格停留时间百分比[雌鼠(8.52±1.93)%,雄鼠(6.95±2.46)%]大于对照组雌、雄大鼠[雌鼠(3.47±0.93)%,雄鼠(4.36±2.22)%](P〈0.05)。消退训练显示,消退训练第1天第一个block音乐组雌、雄大鼠与对照组雌、雄大鼠呆立水平差异无统计学意义;音乐组雌鼠第3天呆立水平显著低于对照组雌鼠(P〈0.05),音乐组雄鼠第2,3天呆立水平均显著低于对照组雄鼠(分别为P〈0.05,P〈0.01)。结论幼龄期音乐暴露有助于改善大鼠创伤应激后的焦虑水平,不影响大鼠的条件恐惧记忆水平,有助于恐惧Objective To study the effect of music exposure in juvenile stage on anxiety-like behavior and fear extinction in adult rats. Methods 4 broods of two-week-old healthy Sprague-Dawleyda rats were separated into two groups randomly. The music groups were exposed to music from 20:00 to 22:00 for 21 consecutive days. When the rats were 5 weeks old,they were caged by sex,with 8 male rats and 6 female rats in each group. When the rats were 8 weeks old, the two groups were assessed in the elevated-plus maze test, the open-field test,trained and assessed by the conditional fear training and fear extinction training. Results In the elevated-plus maze test: Prior to the foot shock,the percent of time spent in the open arms in the music group (female(7.07 ± 1.14)%, male( 5.12 ± 1.95 ) % ) exhibited no significant difference (P〉 0.05 ) comparing with the rats in the control group (female(4.65 ± 0.86 ) %, male (4.86 ± 1.95 ) % ). After the foot shock, the percent of time spent in the open arms in the music group ( female ( 8.63 ± 3.35 ) % , male (7.79 ± 2.49 ) % ) increased comparing to the control group ( female( 1.48 ±0. 11 ) %, male ( 4.29 ± 1.68 ) % ) (P 〈 0.01 ). In the open-field test : prior to the foot shock, the percent of time spent in the center zone of the music group ( female ( 6.16 ±2.17 ) %, male (6.25±3.47 ) % ) exhibited no significant difference (P 〉 0.05 ) comparing with the rats in control group ( female ( 5.27 ±1.95 ) % ,male(6.22 ± 3.13 ) % ). After the foot shock, the percent of time spent in the center zone in the music group ( female( 8.52 ± 1.93 ) %, male ( 6.95 ± 2.46 ) % ) was larger than the control group ( female ( 3.47 ± 0.93 ) % , male (4.36 ± 2.22 ) % ) (P 〈 0.05 ).
