摘  要:目的 探讨镉 (Cd)致淋巴细胞DNA损伤及其镉免疫毒性机制中的地位和作用。方法 运用单细胞凝胶电泳法、MTT法和荧光抗体标记法分别检测各染镉剂量组的DNA损伤、T淋巴细胞增殖转化功能和T淋巴细胞亚群情况。结果 各染镉组的DNA损伤与对照组有显著性差异 (P <0 .0 5 ) ;T淋巴细胞增殖功能与对照比较有降低 (P <0 0 5 ) ,并均随镉剂量的升高有逐步加重的趋势 ;DNA损伤与T淋巴细胞增殖转化功能有显著的相关关系 (Pearson相关分析 ,r =0 .80 78,P <0 .0 1)。结论 镉的免疫毒性机制中镉致淋巴细胞DNA损伤可能是重要的机制之一。Objective To study if the DNA damage of lymphocytes involve in the immunotoxic mechanism of cadmium. Methods The alkaline single cell gel electrophoresis (SCGE, or Comet assay)test, MTT assay and FITC marker were used to study the DNA damage of splenic lymphocyte of rat’s spleen, the proliferation of rat’s splenic lymphocytes, and the subsets of T lymphocyte after the rats had been treated with CdCl 2.Results SCGE showed that DNA damage of splenic lymphocytes in each Cd exposed group were more serious than that of control group with statistical significance ( P <0.05).The proliferation function of T lymphocytes were lower in each Cd exposed group than that in control group ( P <0.05). The decrease of proliferation function seemed to be associated with DNA damage ( P <0.01).Conclusion The result suggested that DNA damage of lymphocytes might be important in the immunotoxic mechanism of cadmium. It would be worth to study further.
