摘  要:目的探讨血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)对心肌营养素-1(cardiotrophin-1,CT-1)的影响及CT-1反义脱氧寡核苷酸、AG490、川芎嗪在CT-1诱导心肌细胞肥大中的作用。方法利用AngⅡ刺激心肌细胞造成细胞肥大、凋亡,应用CT-1反义脱氧寡核苷酸、AG490、川芎嗪进行干预。检测心肌细胞大小及3H-亮氨酸掺入率的变化;以及通过逆转录聚合酶链反应观察CT-1 mRNA表达。结果相差显微镜下可见经AngⅡ刺激的心肌细胞面积变大,3H-亮氨酸掺入率增高;CT-1 mRNA表达增加。CT-1反义脱氧寡核苷酸组心肌细胞面积较肥大组减小,3H-亮氨酸掺入率减少;CT-1 mRNA表达亦减少。AG490和川芎嗪减小心肌细胞面积、3H-亮氨酸掺入率,但不影响CT-1 mRNA的表达。结论 CT-1参与心肌细胞肥大,CT-1反义脱氧寡核苷酸、AG490、川芎嗪可减少心肌细胞肥大。Objective To explore the effect of cardiotrophin-1(CT-1),AG490(an inhibitor of JAK) and tetramethylpyrazine(TMP) in the angiotensin Ⅱ(Ang Ⅱ)-induced myocardial cells hypertrophy.Methods Myocardial cells hypertrophy was induced by Ang Ⅱ stimulation.Then transfection with the CT-1 antisense oligodeoxynucleotide(ASODN) or AG490 and TMP was given to the myocardial cells.Myocardial cells sizes and 3H-leucine incorporation were exmained to evaluate myocardial hypertrophy,and RT-PCR was used to detect CT-1 mRNA expression.Results After Ang Ⅱ stimulation,myocardial cells size,3H-leucine incorporation and CT-1 mRNA expression were increased.Myocardial cells size,3H-leucine incorporation and expression of CT-1 mRNA were decreased in the ASODN group.In the AG490 and TMP group,myocardial cells size and 3H-leucine incorporation were decreased without the effection of the CT-1 mRNA expression.Conclusion CT-1 has relationship with myocardial cells hypertrophy;ASODN,AG490,TMP could inhibit Ang Ⅱ-induced myocardial cells hypertrophy.
