摘  要:目的初步探讨吸入性苯染毒小鼠模型的建立。方法选取50只6~8周龄雌性健康昆明种小鼠,随机分为2组,苯染毒组采用动式吸入式染毒,染毒剂量为2 376.49mg/m3,每天一次,连续染毒21d,空白对照组不处理,实验结束时分别采血进行全血细胞计数,测定脏器指数,骨髓有核细胞计数和微核试验。结果苯染毒组小鼠白细胞、血小板下降为50%(P<0.01),红细胞和血红蛋白显著减少(P<0.05);胸腺、脾指数和肝脏重量明显减小(P<0.01),肾指数增高(P<0.05);骨髓有核细胞显著减少(P<0.01),细胞微核率显著升高(P<0.01)。结论采用动式吸入式染毒,染毒剂量为2 376.49mg/m3,每天一次,连续染毒21d,可初步建立苯染毒小鼠模型。Objective To establish a benzene poisoning mice model by inhalation of benzene in mice.Methods Fifty healthy Kunming(6 to 8 week old)female mice were divided into two groups randomly.Dynamic inhalation of benzene with 2 376.49mg/m3 once daily for 21 days were conducted among the mice in benzene exposure group(control group without additional treatment).The complete blood count,organ indexes of liver,spleen,thymus and kidney,nucleated cells count,micronucleus rate of bone marrow polychromalic erythrocytes(PCE)were detected 24 hour after inhalation of benzene.Results Compared with the control group,the mice in benzene exposure group showed reduction of levels of leukocyte and platelet(50%,P0.01),erythrocyte and hemoglobin(P0.05).The indexes of thymus,spleen index and liver weight were significantly reduced(P0.01),kidney index increased(P0.05).Nucleated cells were significantly reduced(P0.01),micronucleus rate was significantly higher(P0.01).Conclusion A benzene poisoning mice model is established preliminarily by dynamic inhalation of benzene with 2 376.49mg/m3 once daily on 21st day.
