HPLC法测定川产道地药材雅连中盐酸小檗碱的含量

摘　　要：目的:建立川产道地药材雅连中盐酸小檗碱的含量测定方法。方法:采用HPLC法,色谱柱为依利特SinoChrom ODS-BP(250mm×4.6mm,5μm),流动相为乙腈-水(含0.1%乙酸和10mmol/L乙酸铵)梯度洗脱,检测波长345 nm,柱温30℃;流速1.0ml/min;进样量:10μl。结果:盐酸小檗碱在0.25～2.5μg范围内与峰面积线性关系良好(r=0.9999),平均回收率99.29%,RSD为1.38%(n=9)。结论:本方法灵敏、准确、简便快速,可用于川产道地药材雅连的质量控制。Objective：To establish a method for the determination of berberine hydrochloride in Coptis deltoidea C.Y.Cheng et Hsiao in Sichuan.Methods：The separation was carried out on a SinoChrom ODS-BP（250mm×4.6mm,5μm）,the mobile phase was acetonitrile-water（0.1%caproic acid and 10 mmol/L ammonium acetate） with gradient elution,and the detected wavelength was 345nm.The column temperature was 30℃.The flow rate was 1.0 ml/min,and the sample size was 10μl.Results：The calibration curve showed good linearity over the range of 0.25～2.5μg（r=0.9999）.The average recovery was 99.29 % with RSD 1.38%（n=9）.Conclusion：The method is sensitive,accurate,rapid,and it is suitable for the quality control of Coptis deltoidea C.Y.Cheng et Hsiao.
