摘 要:目的建立老年大鼠急性铜绿假单胞菌(PA)肺部感染模型并探讨其炎症反应情况。方法清洁级雄性老年SD大鼠60只随机分为实验组和对照组,实验组采用经气管切开法接种PA标准菌液诱导肺部感染模型,对照组则用等体积灭菌生理盐水替代,动态观察接种PA后2 h、6 h、9 h、12 h、24 h肺组织细菌负荷和病理变化。结果细菌学:实验组大鼠肺组织匀浆菌落计数在接种后2 h、6 h、9 h、12 h均>105cfu/g,接种24 h后>103cfu/g,对照组各时间点均未检出;病理学:实验组大鼠肺组织均有不同程度水肿、充血、纤维化,镜下可见病变较弥散,病灶内有不同程度中性粒细胞(PMN)和单核/巨噬细胞聚集,炎症反应相对轻微,结构破坏较明显,出血、水肿多见,病理改变以6~24 h明显,对照组仅在接种后2~4 h局部有轻微充血水肿、少量炎症细胞浸润,余未见明显变化。结论应用经气管切开法接种PA标准菌液可成功建立老年大鼠急性PA肺部感染模型。Objective To establish an aged rat model of pulmonary infection by inoculating pseudomonas aeruginosa to Sprague-Dawley (SD) rats and to evaluate its inflammatory reaction. Methods Sixty SD rats were divided into 2 groups : the infected group and the control group. The animal model was established through tracheotomy. The bacteria num- ber in the lung and pathological scores were determined 2,6,9,12 and 24 hours after inoculation. Results The bacterio- logical values : no bacterium was detected in the control group. P. aeruginosa was detected from rats of infected groups. Bacte- rial number was more than l05 cfu/g 2,6,9 and 12 hours after infection and more than 103 cfu/g 24 hours after infection. The pathological changes :2,6,9,12 and 24 hours after infection, lung edema, congestion and fibrosis could be seen from lungs of infected groups. Under the optical microscope, in the infected group, there were apparent changes. While in the con- trol group, there were little changes after the inoculation except mild congestion and inflammatory reaction in the lungs 2 and 4 hours after inoculation. Conclusions The animal model of aged rat with pulmonary infection can be established by inoculating pseudomonas aeruginosa through tracheotomy to aged rats.
