摘 要:目的通过寻找与人甘丙肽2型受体(hGalR2)C端相互作用的蛋白,以进一步探讨hGalR2转运和信号传导机制。方法利用酵母双杂交实验寻找可以与hGalR2 C端相互作用的蛋白,并通过酵母双转验证和免疫共沉淀实验验证受体和目标蛋白之间的相互作用。结果酵母双杂交方法结合免疫共沉淀实验发现和证实hGalR2与蛋白Cysteine-rich PDZ-binding protein(CRIPT)之间存在相互作用。结论 CRIPT可以与hGalR2结合而发生相互作用并可能因此参与hGalR2的转运或信号传导。Objective To get insight into the molecular mechanisms of signaling and trafficking of galanin receptor 2 ( GalR2), and to investigate the interaction between human galanin receptor 2 (hGalR2) and cytoplasmic adapter proteins. Methods Yeast two-hybrid method was used to find which proteins interact with the C terminal of hGalR2. Then yeast co- transformation system and co-immunoprecipitation were applied to confirm the protein-protein interaction. Results Cyste- ine-rich PDZ-binding protein (CRIPT) was found interact with the C terminal of hGalR2. The protein-protein interaction was confirmed by yeast co-transformation system and co-immunoprecipitation. Conclusion CRIPT interacts with GalR2 and this protein-protein interaction may be involved in trafficking and signaling of GalR2.
