摘  要:目的:观察愤怒反应模型大鼠顶区皮质和额区皮质5-羟色胺2C受体(5-HTR2C)的表达以及中药经前平颗粒的干预作用,探讨其可能的中枢作用机制。方法:采用居住入侵结合社会隔离的方法制备愤怒反应大鼠模型,并予以经前平颗粒进行药物干预,运用半定量RT-PCR和Western Blot方法检测大鼠顶区皮质和额区皮质中5-HTR2C mRNA及其蛋白表达水平。结果:与正常组相比,愤怒模型大鼠顶区皮质和额区皮质中5-HTR2C mRNA和蛋白表达均显著性升高(P<0.05,P<0.01),经前平颗粒可显著下调其异常表达(P<0.01)。结论:愤怒情志刺激可导致大鼠顶区皮质和额区皮质5-HTR2C表达升高,经前平颗粒可对5-HTR2C表达异常变化起到调节作用,提示5-HTR2C基因与愤怒情绪密切相关,并且可能是经前平颗粒的中枢作用靶点之一。Objective: To explore the central mechanisms of anger emotion and the intervention mechanisms of Jingqianping Granule by observing the changes of the expression level of 5-HTR2C in anger emotional model rats of cerebral cortex regions. Methods: The model rats with anger emotion were prepared by the methods of resident intruder paradigm, and treated with Jingqianping Granule. We used a semi-quantitative RT-PCR gene amplification technology and western blot method to detect the expression levels of 5-HTR2C mRNA and protein in rat cerebral cortex regions. Results: The expression of 5-HTR2C in cerebral cortex of anger emotional model rats increased obviously(P〈0.05, P〈0.01) compared with the normal control group. Compared with the model group, the expression of 5-HTR2C were significantly improved after treated with Jingqianping Graulne(P〈0.01). Conclusion: The anger emotion stimulation obviously increased 5-HTR2C expression in rat cerebral cortex regions. Jingqianping Graulne can obviously reduce its expression. These point out that 5-HTR2C is closely related to anger emotion and the drug-regulating liver recipe might partially plays its role in treating anger emotion by improving the cerebral cortex 5-HTR2C protein and gene expression levels of rats.
