经前平颗粒对愤怒模型大鼠皮质中5-HTR2C表达的干预作用

摘　　要：目的:观察愤怒反应模型大鼠顶区皮质和额区皮质5-羟色胺2C受体(5-HTR2C)的表达以及中药经前平颗粒的干预作用,探讨其可能的中枢作用机制。方法:采用居住入侵结合社会隔离的方法制备愤怒反应大鼠模型,并予以经前平颗粒进行药物干预,运用半定量RT-PCR和Western Blot方法检测大鼠顶区皮质和额区皮质中5-HTR2C mRNA及其蛋白表达水平。结果:与正常组相比,愤怒模型大鼠顶区皮质和额区皮质中5-HTR2C mRNA和蛋白表达均显著性升高(P<0.05,P<0.01),经前平颗粒可显著下调其异常表达(P<0.01)。结论:愤怒情志刺激可导致大鼠顶区皮质和额区皮质5-HTR2C表达升高,经前平颗粒可对5-HTR2C表达异常变化起到调节作用,提示5-HTR2C基因与愤怒情绪密切相关,并且可能是经前平颗粒的中枢作用靶点之一。Objective： To explore the central mechanisms of anger emotion and the intervention mechanisms of Jingqianping Granule by observing the changes of the expression level of 5-HTR2C in anger emotional model rats of cerebral cortex regions. Methods： The model rats with anger emotion were prepared by the methods of resident intruder paradigm, and treated with Jingqianping Granule. We used a semi-quantitative RT-PCR gene amplification technology and western blot method to detect the expression levels of 5-HTR2C mRNA and protein in rat cerebral cortex regions. Results： The expression of 5-HTR2C in cerebral cortex of anger emotional model rats increased obviously（P〈0.05, P〈0.01） compared with the normal control group. Compared with the model group, the expression of 5-HTR2C were significantly improved after treated with Jingqianping Graulne（P〈0.01）. Conclusion： The anger emotion stimulation obviously increased 5-HTR2C expression in rat cerebral cortex regions. Jingqianping Graulne can obviously reduce its expression. These point out that 5-HTR2C is closely related to anger emotion and the drug-regulating liver recipe might partially plays its role in treating anger emotion by improving the cerebral cortex 5-HTR2C protein and gene expression levels of rats.
