摘  要:目的:观察平喘Ⅰ号对呼吸道合胞病毒(RSV)感染引起BALB/c哮喘小鼠肺组织细胞间黏附分子-1(ICAM-1)、血清一氧化氮(NO)含量的影响,探讨其治疗哮喘发作的作用机制。方法:72只BALB/c小鼠随机分为正常对照组、模型对照组、地塞米松组、平喘Ⅰ号低、中、高剂量组6组,每组12只。予RSV致敏,除正常对照组和模型对照组用等量0.9%氯化钠溶液外,其余各组用相应药物干预14d。末次激发48h后取肺组织和血清分别采用免疫组化和ELISA法测定各组ICAM-1和NO含量。结果:与正常对照组比较,模型对照组小鼠ICAM-1、NO含量明显增高(P<0.01)。与模型对照组比较,平喘Ⅰ号各剂量组和地塞米松组均可降低哮喘小鼠ICAM-1和NO水平(P<0.01或P<0.05),其中平喘Ⅰ号高剂量组与地塞米松组水平接近。结论:平喘Ⅰ号能降低哮喘小鼠ICAM-1、NO含量,并具有量效关系。由此推测,通过降低ICAM-1和NO水平,从而减轻气道炎症反应,是平喘Ⅰ号治疗哮喘的作用机制之一。Objective: To observe the effect of Pingchuan I on the levels of lung organization intercellular adhesion molecule-1 (ICAM-1), serum nitric oxide (NO) in respiratory syncytial virus (RSV) infected BALB/c mouse asthma model, and to explore the therapeutic mechanism of asthma attacks. Methods: 72 BALB/c mice were randomly divided into six groups, which were normal control group, model control group, dexamethasone group, Pingchuan Ⅰ low, middle and high-dose group, 12 rats in each. Sensitized to the RSV, in addition to the normal control group and model control group received normal saline, all the other groups with the corresponding drug intervention. 48h after the last challenge, the ICAM-land the NO levels in each group were detected by SABC and ELISA. Results: Compared with normal control group, asthma model groups' ICAM-1, NO were significantly higher (P〈0.01). With the model control group, Pingchuan Ⅰ in each dose group and dexamethasone group could reduce asthma in mice ICAM-1 and NO levels (P〈0.01, P〈0.05), in which, the high dose group and dexamethasone group were similar. Conclusion: The Pingchuan Ⅰ can reduce asthma ICAM-1, NO levels, and has dose-effect relationship. The results suggeste that, by lowering ICAM-1 and NO levels, thereby reducing airway inflammation is the mechanism of Pingchuan I in treating asthma.
