解毒化瘀方对白血病K562/A02细胞耐药逆转作用及NF-κB信号表达的影响

摘　　要：目的:探讨解毒化瘀方中药血清对白血病K562/A02耐药细胞耐药逆转作用及NF-κB信号表达的影响。方法:采用MTT法检测解毒化瘀方中药血清对K562/A02细胞毒作用,流式细胞仪检测解毒化瘀方中药血清对K562/A02细胞内化疗药物浓度的影响,用Western blot法检测解毒化瘀方中药血清对NF-κB/P65、NF-κB/P50、IκBα的表达的影响。结果:解毒化瘀方中药血清对K562/A02细胞生长抑制作用呈浓度依赖性,能够增加K562/A02细胞内化疗药物的浓度,下调K562/A02细胞NF-κB/P65、NF-κB/P50、IκBα表达,从而降低NF-κB信号。结论:解毒化瘀方中药血清能够逆转K562/A02细胞耐药,其耐药逆转机制可能与下调K562/A02细胞NF-κB信号表达有关。Objective： To discuss the influence of Jiedu Huayu formula serum to reverse multidrug resistance and the expression of nuclear factor kappa 13 （NF-κB） in leukemia K562/A02 cell. Methods： MTT assay was adopted to analyze the effect of Chinese herbal serum on growth inhibition in K562/A02 cell and flow cytometry was adopted to analyze the concentration of intracellular DNR. Western blotting was adopted to detect the expression of NF-κB/P65, NF-κB/P50, IκB α in serum which receiving the treatment of Jiedu Huayu formula. Results： Jiedu Huayu formula can inhibit proliferation of K562/A02 cell line, increase the concentration of intracellular DNR. But it also can down-regulate the expression level of NF-κB/P65, NF-κB/PS0, IκB α of k562/A02 cell. Conclusion： Jiedu Huayu formula serum can reverse multidrug resistance in leukemia cell by blocking NF-κB signal pathway of k562/A02 cell.
