“醒脑开窍”针刺法早期介入对脑缺血再灌注大鼠SOD及MDA的影响

摘　　要：目的:观察"醒脑开窍"针法早期介入对脑缺血再灌注模型大鼠超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。方法:将SD成年雄性大鼠随机分为假手术组、模型组、针刺治疗组、药物治疗组,采用线栓法制备大鼠脑缺血再灌注模型,造模成功后,针刺治疗组于造模成功3h后给予"醒脑开窍"针刺法治疗,药物治疗组于造模成功5h后予香丹注射液腹腔内注射。治疗2h后测定血清及脑组织中SOD活性和MDA含量。结果:针刺可提高模型大鼠血清及脑组织SOD活性,且血清MDA有降低趋势,但差异无统计学意义。结论:"醒脑开窍"法早期介入可对SOD、MDA具有一定调节作用,推测针刺对脑缺血再灌注模型大鼠脑组织的保护作用是通过提高SOD活性,抑制脑组织局部脂质过氧化反应,减轻氧自由基对脑组织的损害实现的。Objective： To investigate the effect of early intervention of the ‘Xingnao Kaiqiao＇ acupuncture on the SOD activity and MDA content of the ischemic brain tissues and blood serum at acute period in rats with cerebral ischemia-reperfusion injury. Methods： The male adult SD rats were randomly divided into the sham operation group, the model group, the acupuncture group, the medicine control group. The right side cerebral middle artery of rats was blocked by thread to establish the model of cerebral ischemia-reperfusion. After the model was successfully made for three hours, the acupuncture group was given ‘Xingnao Kaiqiao＇ acupuncture treatment. After the model was successfully made for five hours, the drug group was intraperitoneally injected with Xiangdan injection. Two hours later after the treatment, SOD activity and MDA content was tested in the serum and brain tissue. Results： The acupuncture could increase the SOD activity and decrease MDA content in the the serum and brain tissue of the model group, which had no significant difference. Conclusion： The early intervention of Xingnao Kaiqiao acupuncture has a regulatory role on SOD, MDA, so we suggest that acupuncture has protective effect on the cerebral ischemia-reperfusion of model rats which could be achieved by increasing SOD activity, inhibiting partial lipid peroxidation response in brain tissue and reducing brain tissue damage caused by oxygen free radical.
