摘 要:目的:了解有氧运动对肺动脉高压大鼠肺动脉血管舒张反应的影响。方法:8周大的雄性SD大鼠,随机分为4组,分别是常氧不运动组(N)、常氧运动组(EX)、肺动脉高压不运动组(H)、肺动脉高压运动组(HE)。将大鼠每天暴露在氧浓度为15.6%~16.1%的环境中8小时,每周7次,制作肺动脉高压模型。运动组分别进行8周每天1小时5天/周游泳训练,第9周分离出大鼠肺动脉,检测4组动脉环乙酰胆碱(Ach)诱导的内皮依赖性和硝普钠(SNP)诱导的非内皮依赖性舒张反应。结果:肺动脉高压组大鼠的肺动脉环的内皮依赖性血管舒张功能明显下降,动脉环经NO途径的最大舒张反应明显降低,有氧运动对肺动脉高压大鼠肺动脉的舒张功能有一定的改善作用,与肺动脉高压对照组及常氧运动组比较差异有统计学意义。结论:实验结果证实有氧运动对NO介导的血管舒张功能的有益调节作用,部分被低氧效应所抵消。长期低氧效应超过有氧运动对肺动脉血管的作用这个现象,很可能与NO途径受损有关。Objectives : The purpose of this paper is to investigate the effect of aerobic exercise on vaso- relaxation alterations of pulmonary arteries in pulmonary hypertensive rat. Methods:8 weeks old SD male rats were randomly assigned to 4 groups: normotensive sedentary (N), normotensive trained ( EX), pulmonary hypertensive sedentary ( H), pulmonary hypertensive trained ( HE). Pulmonary hypertension was induced by chronic exposure to hypobaric hypoxia (15.6 - 16.1% oxygen) for eight hours per day (7 days/week). After 56 days, one - hour swimming training per day (5 days/ week), Endothelium- dependent vasorelaxation to acetylcholine (10^-9 - 10^-4M) and endothelium- independent vasorelaxation to sodium nitroprusside (10^10 10^ 4 M) were assessed on isolated pulmo- nary arterial rings. Results : Alterations in endothelium - dependent vasorelaxation properties were ob- tained in pulmonary hypertensive rats. Maximal relaxation to acetylcholine by NO was impaired inboth pulmonary hypertensive groups. Aerobic exercise could affect those pulmonary vasoreactivity al- terations. Conclusion:These results confirm that the potential beneficial effect of exercise on nitric ox- ide mediated pulmonary artery vasorelaxation is partly blunted by deleterious effects of hypoxia. A predominant effect of chronic hypoxia over training seems to be partially responsible for this phenom- enon, probably through impairment in nitric oxide bioavailability.
