摘 要:在过去的几十年中,酶作为经济、环保的生物催化剂已经越来越受到工业以及医药领域的重视,与此同时,高通量筛选方法(HTS)作为酶定向进化中的一项重要技术也得到长足的发展。近些年来,一系列高通量筛选方法的建立使得酶的筛选更加灵敏、高效、快捷,大量的能够适用于工业以及医药领域的工程酶也相继产生。综述了近期建立的一些高通量筛选方法,简要概述了各种筛选方法的基本原理以及存在的优点和不足,并对高通量筛选方法将来的发展做出了展望。During the past decades,the enzyme has gain more and more interest in industry and medicine area as an economic and environmentally friendly bio-catalyst.At the same time,as an important part of directed evolution,HTS methods has made considerable progress.In recent years,a series of high-throughput Screening methods have made the selection of enzyme more sensitive and efficient.Abundant engineered enzymes which can be applied to industry and medicine area have emerged.In this essay,we summarized some newly established high-throughput screening methods,introduced the basic principles,advantages and drawbacks of these screening methods.At last,a prospection of the future developments of high-throughput screening method have also pronounced.
