白花前胡对麻醉开胸猫急性心肌梗塞的影响  

Effects of Peucedanum praeruptorum Dunn, A Traditional Chinese Medicine on Acute Myocradial Infarction in Open chest Anesthetized Cats

摘　　要：目的 :观察中药白花前胡对急性心肌梗塞 (AMI)的影响。方法 :用结扎麻醉开胸猫左冠状动脉前降支诱发 AMI模型 ,于结扎后 10 min经 12指肠给药 ,做冠脉循环及心脏血流动力学 ,心肌耗氧量 ,心外膜电图 ,血清酶活性 ,组织 N- BT染色以及 AMI程度 (Σ- ST)和范围 (N- ST)等检测。结果 :白花前胡醇提物增加冠脉流量 ,降低冠脉阻力、左室压、左室舒张末压、左室最大上升速率、心率、心肌耗氧量、心肌氧摄取率和率压积。给药 (0 .2 g/kg)后 1h,血清乳酸脱氢酶和心肌 MB型肌酸激酶的变化率 ,由对照组的 (6 1.1± 12 .8) %和 (191.1± 2 2 5 .2 ) % ,分别降至(9.1± 44 .6 ) % ,P <0 .0 5和 (- 1.5± 87.2 ) % ;Σ- ST和 N - ST由 (34 .7± 41.2 ) %和 (5 3.6± 8.6 ) % ,分别降至(- 19.0± 15 .4) % ,P <0 .0 5和 (2 2 .2± 4.3) % ,P <0 .0 1;给药后 5 h,左室梗塞率由 (2 3.6± 6 .7) % ,减至 (15 .0±5 .7) % ,P<0 .0 5。结论 :中药白花前胡有抗急性心肌梗塞作用。Objective: To investgate the effects of ethanol extract of Peucedanum praeruptorum Dunn, a traditional Chineses medicinal herb,on acute myocardial infarction(AMI). Methods:Epicardial electrogram,serumal enzymatic assay and histological NBT staining technique were used in open chest urethane anesthetized cats subjected to a 5 h occlusion of the left anterior descending coronary artery. Results:Intraduodenal administration of drug(0.2 g/kg) at 10 min after ligation resulted in a marked increase in CSF,and decreases in CR,BP,LVP,LVEDP,+LV dp/dt max ,MCO 2,MTO 2,RPP and HR. After 1 h of the administration of the drug(0.2 g/kg),LDH and CK MB decreased to(9.1±44.6)% and(-1.5±87.2)%,from (61.1±12.8)% and (191.1±225.2)%,respectively(P<0.05);the severity(Σ ST) and the exten(N ST) of AMI decreased to (-12.6±4.5)%,and (22.2±4.3)%,P<0.01 from(34.7±41.2)% and (53.6±8.6)%,respectively(P<0.5);the percentage of left ventricular infarction deceased to (15.0±5.7)%,from (23.6±6.7)% 5 h after the drug(P<0.05). Conclusion:Peucedanum praeruptorum Dunn shoved a marked cardioprotective effect in AMI cats.
