摘  要:为了筛选出高效培养基,确定适宜的培养方法,以大蒜花序轴为外植体,对大蒜花序轴离体培养诱导鳞茎培养方法进行研究。结果表明,大蒜花序轴离体培养诱导鳞茎最佳培养基为1/2MS+90g·L^-1蔗糖,pH值为5.8~6.0,鳞茎的形成率为96.8%,鳞茎总诱导率为800%。蒜薹总苞表面消毒的最佳方法为75%酒精2min+O.1%升汞5min+O.5%次氯酸钠8min。不添加任何植物激素及添加较高浓度的蔗糖有利于试管鳞茎的形成。The method of bulb induced by garlic rachis in vitro culture was investigated to screen high efficiency culture medium and determine the suitable culture method. The results showed that the optimum medium was 1/2MS+ sucrose 90 g.L^-1, pH 5.8-6.0. The formation rate of bulb was 96.8% and total induction rate of bulblet was 800%.The best method of garlic bolt involucre disinfection was alcohol 75% (2 min)+ mercuric chloride 0.1% (5 min) + sodium hypochlorite 0.5% (8 min). Adding no hormone and high concentration sucrose was advantageous to formation of bulb in vitro.
