摘 要:为了进行黄豆酱中低聚肽的分离,去除黄豆酱中的盐分。以脱盐率、肽吸附率为指标,对黄豆酱脱盐的吸附树脂种类、动态吸附及解吸条件进行了研究。筛选出了强酸性苯乙烯系阳离子交换树脂001×7为吸附剂,动态吸附的最佳脱盐条件为进样浓度4 mg.mL-1,pH为8的50%乙醇溶液为洗脱剂,流速30 r.min-1,在此条件下,脱盐率为91.28%,肽回收率为88.54%。研究表明,采用001×7阳离子交换树脂对黄豆酱脱盐是一种有效的方法。本试验为黄豆酱多肽的进一步分离、生物活性评价及其一级结构鉴定奠定了研究基础。Soybean paste was desahed for separating the oligopeptides by using the method of desalination dynamic adsorption and desorption. The desalination rate and peptide adsorption rate were used to analysis the effect of desalination conditions of soybean paste. The results showed that the 001 ×7 strong acid styrene cation exchange resin was appropriate adsorbent, the optimum desalination condition as follows: loading sample concentration was 4 mg'mL^-1, the pH=8 of 50% alcohol was eluent, the flow speed was 30 r.min^-1. Optimally, the ratio of desalination reached to 91.28%, and the recovery of peptides was 88.54%. This study suggested that desalination method by using 001 ×7 cation exchange resin adsorption resin applied to soybean paste was an available method. This study provided experimental foundation for further research about the separation, activity and the identification primary structure of soybean paste peptides.
